牙龈成纤维细胞诱导分化神经细胞的可行性 被引量:1

摘要 目的 探讨牙龈成纤维细胞诱导分化神经细胞的可行性。方法 选取阻生齿拔除患者10例,取其周围正常牙龈组织作为研究样本,分析牙龈成纤维细胞诱导分化神经细胞的可行性。结果 人牙龈成纤维细胞有突起,胞体呈纺锤形或梭形,存在明显细胞轮廓,核呈卵圆形,细胞体及细胞核均较大,有明显核仁,着色浅;人牙龈成纤维细胞经染色显示为vimentin表达阳性,胶质纤维酸性蛋白(GFAP)、人微管相关蛋白(MAP)-2表达阴性;神经样细胞呈三角形或星形,存在3个及以上突起,细胞核变圆,突起细长,出现轴突样和树突样结构,且互相连接,呈现网状结构;经染色显示神经样细胞为vimentin表达、GFAP表达、MAP-2表达均为阳性;诱导组乙酰胆碱及多巴胺含量均明显高于对照组(均P<0.05),诱导组乙酰胆碱及多巴胺浓度均明显高于对照组(均P<0.05)。结论 对牙龈成纤维细胞进行诱导处理可获得神经样细胞,诱导后细胞不仅具有神经细胞结构、形态,同样具有神经细胞的分泌功能,分泌大量的乙酰胆碱及多巴胺等神经递质。 Objective To explore the feasibility of gingival fibroblast-induced differentiation of nerve cells.Methods 10 patients with impacted tooth extraction were selected, the normal gingival tissue around them was taken as a research sample to analyze the differentiation of gingival fibroblast-induced neural cells.Results Human gingival fibroblasts had protrusions, and the cell body was spindle-shaped or fusiform.There was a clear cell outline, and the nucleus was oval.The cell body and nucleus were large, with obvious nucleolus and light color.The staining showed positive expression of vimentin in human gingival fibroblasts, negative expression of glial fibrillary acidic protein(GFAP) and human microtubule-associated proteins(MAP)-2;nerve-like cells were triangular or star-shaped, with three or more protrusions, the nucleus became round, the protrusions were elongated, and axon-like and dendritic-like structures appeared which connected to each other, showing a network structure;staining showed that neuronal cells were positive for vimentin, GFAP and MAP-2 expression;the acetylcholine content and dopamine content in induction group were significantly higher than those in control group(all P<0.05).The concentration of acetylcholine and dopamine in induction group were higher than those in control group(all P<0.05).Conclusions Induction treatment of gingival fibroblasts could obtain nerve-like cells.After induction, the cells not only have the structure and morphology of nerve cells, but also have the secretion function of nerve cells, secreting a large amount of neurotransmitters such as acetylcholine and dopamine.
