摘要
通过形态学和分子生物学鉴定一株源自山羊,疑似烟曲霉(命名为YQM6)的菌株,并研究其致病性和耐药性.PCR扩增其毒力基因,通过小鼠致病性试验和药敏试验探究该分离株特性.试验结果显示,YQM6菌株与GenBank中登录的烟曲霉(MZ150 487)同源性为100%,在系统进化树中与烟曲霉(NR121 481.1)聚为一支.扩增得到601 bp ITS基因片段,检测出小G蛋白(RHO1)、蛋白激酶A调节亚基(PkaR)、过氧化氢酶1(Cat1)、碱性蛋白酶(Alp2)、核糖核酸酶(AspF1)等5个毒力基因.试验小鼠见肝、肾肿大,肺及肠有不同程度的出血,心肌血管充血,肝细胞体积增大、水肿,脾小体淋巴细胞减少,肺毛细血管扩张.该菌株对伊曲康唑、伏立康唑敏感,对卡泊芬净耐药.综上,YQM6菌株被鉴定为烟曲霉,且该菌株具有一定致病性.
Identification by morphology and molecular biology of a goat-derived strain of Aspergillus fumigatus(YQM6)and study of its pathogenicity and drug resistance. The virulence genes of YQM6 strain were amplified by PCR.The characteristics of the isolated strains were explored by drug sensitivity test and mouse pathogenicity test.The experimental results showed that YQM6 strain has 100 % homology with Aspergillus fumigatus(MZ150487),which is registered in GenBank.In the phylogenetic tree YQM6 strain ctusters with Aspergillus fumigatus(NR121481.1) as a group.The 601 bp ITS gene fragment was amplified by amplification.Five virulence genes were detected, including small G protein(RHO1),protein kinase A regulatory subunit(PkaR),peroxidase 1(Cat1),alkaline protease(Alp2) and ribonuclease(AspF1).Anatomical test mice exhibited liver and renal enlargement.There were different degrees of bleeding in the lungs and intestines.HE staining showed congested myocardial vessels, enlarged hepatocytes with oedema, reduced lymphocytes in splenic vesicles, and dilated capillaries in the lungs.Drug sensitivity tests showed that YQM6 strain was sensitive to itraconazole and resistant to caspofungin.In conclusion, the YQM6 strain is identified as Aspergillus fumigatus,and the strain has certain pathogenicity.
