摘要
目的探讨CD44在结肠癌组织及结肠癌细胞中的表达意义。方法收集浙江省舟山医院2021年1-12月收治的56例结肠癌患者结肠癌组织及癌旁组织,采用二氨基联苯胺(DAB)法测定并比较结肠癌组织及癌旁组织的细胞CD44阳性率,比较不同病理特征患者结肠癌组织细胞CD44阳性率;采用异硫氰酸酯(FITC)染色法判定结肠癌细胞系HT29及正常结肠上皮细胞系NSM460的CD44阳性率,采用Western blot法测定并比较两组细胞CD44蛋白表达水平。结果结肠癌组织细胞CD44阳性率(66.07%)高于癌旁组织(14.28%),差异有统计学意义(P<0.05)。不同性别、年龄和肿瘤直径癌组织细胞CD44阳性率比较,差异无统计学意义(P>0.05);C~D期癌组织细胞CD44阳性率高于A~B期癌组织,低分化癌组织细胞CD44阳性率高于中、高分化癌组织,淋巴结转移患者癌组织细胞CD44阳性率高于无淋巴结转移患者癌组织,差异均有统计学意义(均P<0.05)。FITC染色结果显示,HT29的CD44阳性率高于NSM460,差异有统计学意义(P<0.05)。Western blot结果显示,HT29中CD44蛋白灰度值高于NSM460,差异有统计学意义(P<0.05)。结论CD44在结肠癌组织及结肠癌细胞中均高表达。
Objective To investigate the expression of CD44 and its significance in colon cancer tissue and cells.Methods The samples of cancer tissues and adjacent tissues of 56 patients with colon cancer treated in Zhejiang Zhoushan Hospital from January 2021 to December 2021 were collected.The positive rate of CD44 was determined by immunohistochemistry DAB method in tissue samples;and determined in colon cancer cell line HT29 and normal colon epithelial cell line NSM460 by FITC staining.The positive rate of CD44 was compared between colon cancer tissues and adjacent tissues,among colon cancer tissues of different pathological types,and between HT29 and NSM460 cells.Results The positive rate of CD44 in colon cancer tissues was significantly higher than that in adjacent tissues(66.07%vs.14.28%,P<0.05).There was no significant difference in the positive rate of CD44 among patients with different sexes,ages and tumor diameters(P>0.05).The positive rate of CD44 in stage C-D cancer tissues was higher than that in stage A-B cancer tissues,in poorly differentiated cancer tissues was higher than that in moderately and highly differentiated cancer tissues,in colon cancer with lymph node metastasis was higher than that without lymph node metastasis(all P<0.05).FITC staining showed that the positive rate of CD44 in HT29 cells was higher than that in NSM460 cells(P<0.05).Western blot results showed that the gray value of CD44 protein in colon cancer HT29 cells was higher than that that in normal colon epithelial NSM460 cells(P<0.05).Conclusion CD44 is highly expressed in colon cancer tissues and colon cancer cells.
