摘要
目的研究宣痹通瘀胶囊对心肌缺血再灌注损伤(MIRI)大鼠的保护作用。方法50只大鼠随机分为5组(n=10):假手术组、模型组、宣痹通瘀胶囊组、宣痹通瘀+LY294002组以及阳性对照药通心络胶囊组。各组动物每日给药1次,连续2周。末次给药后实施肋间开胸手术,结扎冠状动脉左前下降支,缺血30 min后松解结扎线进行再灌注2 h,建立NAFLD大鼠动物MIRI模型。检测指标:血流动力学指标LVSP、LVEDP、±dp/dtmax;血清酶学指标CK-MB、LDH、SOD、cTnI活性以及MDA含量;检测心肌梗死面积;检测心肌缺血模型大鼠心肌细胞凋亡指数。结果宣痹通瘀胶囊3.2 g/kg剂量组大鼠血清中LDH、cTnI、CK-MB含量显著降低;可明显升高LVSP、+dp/dtmax;可以明显减小大鼠心肌梗死范围;大鼠心肌细胞凋亡率有明显改善;降低MDA含量;升高SOD活性、分别与MIRI模型组比较,P<0.01或P<0.05。结论宣痹通瘀胶囊预防性给药后,可抑制心肌细胞凋亡,减轻心肌损伤,对心肌缺血再灌注损伤具有保护作用。
Objective To study the protective effect of Xuanbi Tongyu Capsules on the myocardial ischemiareperfusion injury(MIRI)in rats.Methods Fifty rats were randomly divided into 5 groups(n=10):sham operation group,model group,Xuanbi Tongyu Capsules group,Xuanbi Tongyu Capsules plus LY294002 group and Tongxinluo Capsules positive control group.The subjects in each group were given the corresponding treatment once a day for two consecutive weeks.After the last drug administration,an intercostal thoracotomy was performed,and the left anterior descending coronary artery was ligated.After 30 minutes of ischemia,the ligature was released and a reperfusion was performed for 2 hours to establish the MIRI model in the NAFLD rats.The hemodynamic indexes LVSP,LVEDP and±dp/dtmax,the serum enzyme indexes CK-MB,LDH,SOD,cTnI activity and the MDA content,the size of myocardial infarction were measured.The apoptosis index of myocardial cells was detected in the myocardial ischemia model rats.Results The contents of LDH,cTnI and CK-MB in the rats serum in the Xuanbi Tongyu Capsules group(with a 3.2 g/kg dose)were signifi cantly decreased,while the levels of LVSP and+dp/dtmax were signifi cantly increased.The myocardial infarction area was signifi cantly reduced and the apoptosis rate of myocardial cells in the rats was signifi cantly improved.The MDA content was reduced while the SOD activity was increased respectively by P<0.01 and P<0.05 compared with the MIRI model group.Conclusion After a prophylactic administration,Xuanbi Tongyu Capsules can inhibit the myocardial cell apoptosis,alleviate the myocardial injury,which has a protective eff ect on the myocardial ischemia-reperfusion injury in a MIRI rat model.
