摘要
转基因玉米MON87419是耐受麦草畏和草铵膦两种除草剂的玉米新品系。为了建立转基因玉米MON87419实时荧光定量检测方法,本研究根据转基因玉米MON87419的3′端旁侧序列信息设计引物和Taqman探针,经引物筛选、PCR体系优化、特异性检测、标准曲线建立、正确度及精密度检测和定量限测试等试验,表明该方法能特异、定量检测出转基因玉米MON87419转化体成分,可以对0.1%以上的MON87419转基因成分进行准确定量。本研究结果可为该转基因玉米品系的安全监管提供有效技术支撑。
Genetically modified(GM)maize MON87419 is a new herbicide-tolerant maize line to Dicamba and Glufosinate.In order to establish a real-time fluorescence quantitative PCR detection method for GM maize MON87419,primers and TaqMan probes were designed based on its 3′side sequence information,and the primer screening,PCR system optimization,specificity detection,standard curve establishment,correctness and precision detection and limit of quantitative(LOQ)were conducted.The results showed that the established method could detect the GM maize MON87419 specifically and quantitatively,and more than 0.1%of MON87419 transgenic components could be accurately quantified.This method could provide effective technical support for safety supervision of the new GM maize.
