摘要
建立了高效液相色谱法(HPLC)测定奶制品中苯甲酸、山梨酸、糖精钠、甲基香兰素和乙基香兰素的方法。样品经磷酸盐缓冲溶液和甲醇(V∶V=85∶15)直接提取,乙酸锌-亚铁氰化钾体系沉淀蛋白质,PXA固相萃取小柱净化,外标法和溶剂标准曲线定量。结果表明,5种食品添加剂在0.5~20.0μg·mL^(-1)浓度范围内呈良好线性,相关系数均大于0.9997,检出限均为1.00 mg·kg^(-1),定量限均为3.00 mg·kg^(-1)。3个不同添加浓度(5.0、10.0、50.0 mg·kg^(-1))的平均回收率在80.0%~100%之间,相对标准偏差(RSD,n=6)在0.40%~7.4%之间。
A method for the determination of benzoic acid,sorbic acid,saccharin sodium,methyl vanillin and ethyl vanillin in dairy products by High performance liquid chromatograph.The samples were directly extracted by phosphate buffer solution and methanol(V∶V=85∶15),protein precipitated by zinc acetate and potassium ferrocyanide system,purified by PXA solid phase extraction column,qualitative and quantitative by external standard method and solvent standard curve.The results showed that the five food additives showed good linearity in the concentration range of 0.5-20.0μg·mL^(-1),the correlation coefficients were greater than 0.9997,the limits of detection were1.00 mg·kg^(-1),and the limits of quantification were 3.00 mg·kg^(-1).The average recoveries of the three concentrations(5.0,10.0 and 50.0 mg·kg^(-1))ranged from 80.0%to 100%,and the relative standard deviations(RSD,n=6)ranged from 0.40%to 7.4%.
