摘要
目的建立白花蛇舌草中齐墩果酸和熊果酸的高效液相测定方法,并采用正交设计法优化白花蛇舌草提取工艺。方法色谱柱:Aglient C_(18)(4.6 mm×150 mm,5μm);流动相:乙腈-磷酸盐缓冲液pH=6.8(70∶30);检测波长:210 nm;流速:0.8 mL·min^(-1);柱温:25℃。结果齐墩果酸的线性范围在0.1624~1.2992μg,r=0.9992,平均回收率为102.22%,RSD为0.83%;熊果酸线性范围在0.3296~2.6368μg,r=0.9991,平均回收率为100.78%,RSD为0.46%。齐墩果酸和熊果酸平均加样回收率(n=9)分别为102.2%、100.7%,RSD均不大于1.2%。最佳提取工艺为乙醇浓度为90%,料液比1∶80,超声提取30 min,超声功率120 W。结论本实验方法操作简便,定量准确,重现性好,可用于白花蛇舌草药材中齐墩果酸、熊果酸的含量测定。
Objective To develop HPLC method for determination of oleanolic acid and ursolic acid in Hedyotis diffusa Willd.,and to optimize the extraction technology with orthogonal design.Methods Chromatographic column:C18 Aglient(150 mm×4.6 mm,5μm),mobile phase:acetonitrile-phosphate buffer pH=6.8(70∶30),detection wavelength:210 nm,flow rate:0.8 mL·min^(-1),column temperature:25℃.Results The linear range of oleanolic acid in 0.0406~1.2992μg,r=0.9992.The average recovery rate for 102.22%,RSD was 0.83%;ursolic acid linear range in 0.0824~2.6368μg,r=0.9991.The average recovery rate for 100.78%and the RSD was 0.68%.The average recoveries of oleanolic acid and ursolic acid(n=9)were 102.2%,100.7%and RSD were no more than 1.2%,respectively.The best extraction process is 80 times the amount of 90%ethanol,ultrasonic extraction of 30 min,ultrasonic power of 120 W.Conclusion The method is simple,accurate and with good reproducibility.It can be used to determine the content of oleanolic acid and ursolic acid in Hedyotis diffusa Willd.
