摘要
【目的】分析和田羊Lin28B基因的启动子序列与结构,为进一步解析其转录调控机制奠定基础。【方法】根据Ensembl数据库中绵羊Lin28B基因5’侧翼区序列,设计PCR引物进行扩增、克隆测序及序列鉴定,得到和田羊Lin28B基因启动子区序列,并利用相关生物信息学软件对其进行预测分析。【结果】试验成功获得2 993 bp的和田羊Lin28B基因启动子区序列,GC含量为39.33%;预测了其核心启动子区域,发现在启动子区存在7个潜在的转录起始位点,同时发现了2个TATA-box、2个CpG岛,但未发现CAAT-box;该序列还包含AP-1、Oct-1、p53、Pax-4、STAT3、TGIF等转录因子结合位点。【结论】运用生物信息学软件预测分析和田羊Lin28B基因启动子序列,可为更深入研究Lin28B基因对绵羊初情期的调控机制提供理论基础。
[Objective] The promoter sequence and structure of Lin28 B gene in Hetian sheep were analyzed to lay the foundation for further analysis of its transcriptional regulation mechanism.[Method] According to the 5’ flanking region sequence of sheep Lin28 B gene in Ensembl database,PCR primers were designed for amplification,cloning,sequencing and sequence identification to obtain the promoter region sequence of Lin28 B gene in Hetian sheep, and the relevant bioinformatics softwares were used to predict and analyze the promoter region sequence.[Result] The 2 993 bp sequence of Lin28 B gene promoter region of Hetian sheep was successfully obtained, and GC content was 39.33%.The core promoter region was predicted,and 7 potential transcription start sites were found in the promoter region.Besides, 2 TATA-boxes and2 CpG islands were found, but no CAAT-box was found.The sequence was also found to contain binding sites for transcription factors such as AP-1,Oct-1,p53,Pax-4,STAT3 and TGIF.[Conclusion] Bioinformatics softwares were used to predict and analyze the promoter sequence of Lin28 B gene in Hetian Sheep, which can provide a theoretical basis for further research on the regulation mechanism of Lin28 B gene on sheep puberty.
