摘要
试验应用反向高效液相色谱仪测定饲料原料中色氨酸的含量。色氨酸样品经过碱水解处理,以乙酸钠缓冲液和甲醇为流动相,在280 nm的紫外波长下进样检测。实验结果显示,色氨酸标准溶液浓度在6.977~558.195 mg/L范围内,浓度与峰面积线性关系良好(R^(2)=0.9996)。经过检测分析,该方法的相对标准偏差RSD为0.69%,样品加标回收率为90.15%~105.39%,方法检出限为17.32 ng/kg,定量限为191.22 ng/kg。该实验方法简单可靠,能准确测定饲料原料中的色氨酸。
The tryptophan in feed ingredients was determined by reverse high performance liquid chromatography.The samples were hydrolyzed by alkaline hydrolysis.Tryptophan was determinated by HPLC with UV detection at 280 nm,and the mobile phase was sodium acetatesolution and methanol.The results showed that when the concentration of tryptophan standard solution was in the range of 6.977~558.195 mg/L,and the linear relationship between concentration and peak area was good(R^(2)=0.9996).After detection and analysis,the relative standard deviation RSD of the method was 0.69%,the recoveries were 90.15%-105.39%,the detection limit of the method was 17.32 ng/kg,and the quantification limit was 191.22 ng/kg.This method is simple and reliable,and can be used for the detection of tryptophan in feed ingredients.
