基于SRAP技术的地被竹资源遗传多样性分析 被引量:1

Genetic Diversity Analysis of Dwarf Bamboo Resources Based on SRAP Technology

摘要 地被竹生长低矮紧凑,具有搭配景观效果,在园林植物应用中具有独特作用,因分布较广,遗传背景较为复杂,开展遗传多样性分析对竹类种质资源保护具有重要的现实意义。本研究利用SRAP标记对35个竹种进行遗传多样性分析和鉴定,10对SRAP引物共产生169条谱带,平均每个引物组合可扩增出16.9条谱带,其中多态率为100%,me1/em14、me2/em5、me3/em4和me5/em34个引物组合可区分31个竹种。聚类结果显示,在遗传相似系数0.146处,35个竹种可以分为3大类群,第Ⅰ类群包括‘白纹椎谷笹’、‘鹅毛竹’、‘菲白竹’、‘菲黄竹’、‘银丝竹’、‘黑竹’、‘翠竹’和‘观音竹’,第Ⅱ类群包括‘铺地竹’、‘无毛翠竹’、‘小琴丝竹’、‘凤尾竹’、‘黄条金刚竹’和‘芦花竹’,第Ⅲ类群包括21个竹种,在遗传相似系数0.226处又分为4亚类。研究结果表明地被竹不同种之间亲缘关系较远,遗传背景较复杂。 The dwarf bamboo grows low and compact and plays a unique role in the application of garden plants with landscape effect.Due to its wide distribution and complex genetic background,the analysis of genetic diversity has important practical significance for the protection of bamboo germplasm resources.In this study,SRAP markers were used to analyze and identify the genetic diversity of 35 bamboo species.The results showed that 10pairs of SRAP primers produced a total of 169 bands,and each primer combination amplified an average of 16.9bands with polymorphism rate 100%,indicating that the polymorphism of SRAP sites detected among 35 bamboo species was high,and the four primer combinations of me1/em14,me2/em5,me3/em4 and me5/em3 could distin guish 31 bamboo species.Clustering results showed that the genetic similarity coefficient of 0.146,35 bamboo species could be divided into three groups,the 1 group including‘Baiwenzhuiguti’,‘Emaozhu’,’Feibaizhu’,‘Feihuangzhu’,‘Yinsizhu’,‘Heizhu’,‘Cuizhu’and‘Guanyinzhu’,the Ⅱ groups including‘Pudizhu’,‘Wumaocuizhu’,‘Xiaoqinsizhu’,‘Fengweizhu’,‘Damingzhu’,‘Huangtiaojingangzhu’and‘Luhuazhu’,the Ⅲ groups including 21 bamboo species and dividing into four subgroups.The results showed that the relationship between different species of dwarf bamboo was far and the genetic background was complex.
