摘要 为了减少畜禽粪便堆肥过程中NH_(3)的释放、降低氮损失,文章研究运用富集驯化、初筛复筛的方式从微生物菌剂中筛选出一株除氨菌株ZYC-1,通过形态学、16S r RNA基因序列分析和生理生化鉴定,初步确定ZYC-1为克雷伯氏菌(Klebsiella sp.)。对ZYC-1进行培养条件的优化,通过考马斯亮蓝法测定24 h时的胞内粗酶(亚硝酸盐氧化还原酶(NXR)、亚硝酸还原酶(NIR)、硝酸盐还原酶(Nar)等)浓度,确定ZYC-1在1.0%接菌量、初始pH 7.0、180 r/min、30℃条件下生长最好。在除氨特性的研究中,通过测定NH_(4)^(+)、NH_(2)OH、NO_(2)^(-)、NO_(3)^(-)以及总氮(TN)和溶解性有机碳(DOC)的含量变化发现,在24 h内NH_(4)^(+)、TN、DOC的降解率分别为98.51%、97.01%、79.60%,说明ZYC-1可以高效降解NH_(4)^(+)和TN。分别测定NXR、NIR和Nar 3种关键酶时发现均有表达量,推测硝化-反硝化途径同时存在,表明ZYC-1在畜禽粪污除氨保氮领域具有潜在的利用价值。 In order to reduce the release of NH_(3) and the loss of nitrogen during the composting of livestock and poultry manure,an ammonia-removing strain ZYC-1 was screened from microbial agents by means of enrichment,domestication,primary screening and rescreening.Through morphology,16S rRNA gene sequence analysis and physiological and biochemical identification,it was preliminarily determined that ZYC-1 was Klebsiella sp.The culture conditions of ZYC-1 were optimized,and the intracellular crude enzymes(nitrate oxidoreductase(NXR),nitrite reductase(NIR),nitrate reductase(Nar),etc.)at 24 h were determined by Coomassie brilliant blue method.The concentration of ZYC-1 was determined to grow best under the conditions of 1.0%inoculum,initial pH 7.0,180 r/min,and 30℃.In the study of ammonia removal characteristics,by measuring the content changes of NH_(4)^(+),NH_(2)OH,NO_(2)^(-),NO_(3)^(-),total nitrogen(TN)and dissolved organic carbon(DOC),it was found that the degradation rates of NH_(4)^(+),TN and DOC within 24 h were 98.51%,97.01%,79.60%,respectively,indicating that ZYC-1 can efficiently degrade NH_(4)^(+)and TN.It is speculated that the nitrification denitrification pathway exists at the same time,indicating that ZYC-1 has potential utilization value in the field of ammonia removal and nitrogen conservation of livestock and poultry manure.
