极小种群馨香玉兰特异性单核苷酸多态性位点开发

Development of SNPs in Plant Species with Extremely Small Populations of Magnoliaodoratissima Based on SLAF-seq Technique

摘要 在馨香玉兰全基因组范围内进行特异性单核苷酸多态性(SNP)位点开发,为馨香玉兰濒危保护以及资源利用提供理论依据。利用以云南省5个野生居群和2个栽培居群共70个个体为材料,进行简化基因组测序技术(SLAF-seq),成功开发了843082个用于馨香玉兰SLAF建库的标签,其中多态性SLAF标记有287843个,通过数据质控,共获得了180650个高度一致性的群体SNP位点,利用SNP位点对馨香玉兰居群进行遗传多样性分析,结果显示:馨香玉兰的观测杂合度H_(o)为0.2697,期望杂合度H_(e)为0.3355,Shannon指数为0.5069,Nei多样性指数为0.3553。结果表明,利用SLAF-seq技术能够实现全基因组范围内的大量SNP标记位点开发,利用大量的SNP位点解析物种的遗传多样性更具准确性和先进性。 In this study,specific SNP molecules were developed and labeled in the whole genome of Magnolia odoratissima population,providing theoretical basis for endangered species protection and resource utilization.Using the simplified genomesequencing technique(SLAF-seq),a total of 70 individuals of M.odoratissima from five wild populations and two cultivated populations in Yunnan province were selected.Single nucleotide polymorphism(SNP)markers with high stability and specificity have been developed on the polymorphic SLAF tag.A total of 843082 tags were successfully developed for SLAF library construction and 287843 SLAF markers was polymorphic.Through data quality control,180650 highly consistent population SNPs were obtained.The genetic diversity of the population of M.odoratissima was analyzed using SNP loci,and the results showed that:the observed heterozygosity H_(o) was 0.2697,the expected heterozygosity H_(e) was 0.3355,Shannon’s wiener index was 0.5069,Nei’s diversity index was 0.3553.The results indicate that SLAF-seq can be used to develop a large number of SNP markers in the whole genome,and it is more accurate and advanced by using SNP markers to analyze the genetic diversity of species.
