摘要
沉默信息调节因子2相关酶1(SIRT1)是烟酰胺腺嘌呤二核苷酸(NAD+)依赖性脱乙酰酶Sirtuin家族成员,SIRT1与神经退行性疾病、心脑血管疾病、脂肪肝和肿瘤性疾病等炎性疾病密切相关。本文主要综述了SIRT1的分子调控机制及其在炎性疾病中的作用,具体介绍了SIRT1及其相关下游转录因子肿瘤蛋白53(p53)、核因子-κB(NF-κB)、单磷酸腺苷依赖的蛋白激酶(AMPK)、过氧化物酶体增殖物激活受体γ辅激活因子-1α(PGC-1α)等分子的调控机制以及SIRT1在相关疾病中的作用,以期为多种炎性疾病的治疗提供技术支撑。
Silent information regulator 2-related enzyme 1(SIRT1)is a member of the Sirtuin family of nicotinamide adenine dinucleotide(NAD+)dependent deacetylases.SIRT1 is closely related to inflammatory diseases such as neurodegenerative diseases,cardiovascular and cerebrovascular diseases,fatty liver and tumor diseases.The regulatory mechanism among the SIRT1 and tumor protein 53(p53),nuclear factor-κB(NF-κB),adenosine monophosphate-activated protein kinase(AMPK)and peroxisome proliferator-activated receptorγcoactivator-1α(PGC-1α),and the role of SIRT1 in inflammation were reviewed in this article,in order to provide technical support for the treatment of various inflammatory diseases.
