摘要 【目的】为探究板栗花粉萌发培养基以及乙二醇二乙醚二胺四乙酸对板栗花粉管发育的影响,明确Ca^(2+)在花粉萌发中的作用。【方法】以处于盛花期的‘燕山红栗’花粉为试材,首先筛选花粉萌发培养基配方,随后以此为基础添加不同浓度的乙二醇二乙醚二胺四乙酸,观察乙二醇二乙醚二胺四乙酸对花粉萌发和花粉管伸长以及纤维素和胼胝质分布的影响。【结果】板栗花粉最适宜离体萌发培养基为7.5%蔗糖、0.0010%H_(3)BO_(3)、0.004%CaCl_(2)和1%琼脂,34℃培养36 h,此时花粉萌发率最高为34.01%;添加乙二醇二乙醚二胺四乙酸后,花粉管伸长受阻,当乙二醇二乙醚二胺四乙酸浓度为800μmol/L时,花粉管顶端出现较多的纤维素和胼胝质沉积。【结论】乙二醇二乙醚二胺四乙酸抑制花粉萌发及花粉管伸长,同时导致花粉管的纤维素和胼胝质分布异常,该研究为揭示板栗花粉响应胁迫的应答机制及花粉萌发和花粉管发育机理提供新的思路。 【Objective】This study aims to optimize condition for pollen germination and investigate ethylene glycol tetraacetic acid on pollen tube development of Castanea mollissima and clarify the role of Ca^(2+)in pollen germination.【Methods】In this study,the pollen of‘Yanshanhongli’at full bloom was collected as materials.Firstly,the medium was selected for pollen germination.Then different concentrations of ethylene glycol tetraacetic acid were added into the medium,and the effects of ethylene glycol tetraacetic acid on pollen germination,pollen tube growth,and the distribution of cellulose and callose in pollen tube wall were investigated.【Results】The most suitable solid medium for pollen germination contained 7.5%sucrose,0.0010%H_(3)BO_(3),0.004%CaCl_(2),and 1%agar,on which the pollen exhibited the highest germination rate(34.01%)after culturing for 36 h at 34℃.After addition of ethylene glycol tetraacetic acid,the growth of pollen tube was inhibited.When ethylene glycol tetraacetic acid concentration was 800μmol/L,a lot of cellulose and callose deposit appeared at the top of pollen tube.【Conclusion】Ethylene glycol tetraacetic acid inhibited pollen germination and pollen tube growth,and resulted in the abnormal distribution of cellulose and callose at the top of pollen tube.These results provided new evidence for the response mechanism of pollen to stress,and pollen germination and pollen tube development of Castanea mollissima.
