基于网络药理学探究芪药消渴胶囊治疗2型糖尿病的作用机制 被引量:2

Exploring the Mechanism of Qiyao Xiaoke Capsule(芪药消渴胶囊)in Treating Type 2 Diabetes Based on Network Pharmacology

摘要 目的借助网络药理学探究芪药消渴胶囊治疗2型糖尿病的作用机制。方法通过中药系统药理学分析平台(TCMSP)和中医药百科全书数据库(ETCM)筛选芪药消渴胶囊中十二味中药潜在靶点信息,通过人类基因数据库(Gen-ecards)搜索T2DM疾病的相关靶点信息,将二者取交集得到成药-疾病交集靶点,制作维恩图(Venn);使用STRING平台构建交集靶点的PPI网络,将PPI网络TSV文件导入Cytoscape3.7.1软件,运用Cytoscape3.7.1软件进行关键靶点分析,根据Degre值进行二次构建,做出明暗大小变化,筛选出关键核心靶点;借助Metascape数据库进行GO和KEGG富集分析,筛选出芪药消渴胶囊治疗2型糖尿病的主要活性成分。结果获得药物251个潜在靶点,疾病1362个潜在靶点,二者交集靶点140个,筛选后的核心靶点包括MAPK类蛋白、白细胞介素-6(IL-6)、AKT通路、肿瘤坏子因子(TNF)等;参与的生物进程包括活性氧代谢过程、氧水平反应、脂多糖反应、凋亡信号通路、细胞核受体活性、血红素结合作用、炎症反应等;相关的通路包括:AGE-RAGE信号通路在糖尿病并发症中的作用、蛋白聚糖在癌症中的作用、NF-κB信号通路、鞘脂信号通路、胰岛素抵抗通路等。结论通过网络药理学的进一步探索,发现芪药消渴胶囊的活性成分是以多靶点、多基因、多通路治疗2型糖尿病的作用机制。 Objective To explore the mechanism of action of Qiyao Xiaoke Capsule(芪药消渴胶囊)in the treatment of type 2 diabetes with the help of network pharmacology.Methods The potential target information of twelve herbs in Qiyao Xiaoke Capsule was screened by Systematic Pharmacological Analysis Platform of Traditional Chinese Medicine(TCMSP)and Encyclopedia of Traditional Chinese Medicine Database(ETCM),the relevant target information of T2DM disease was searched by Human Gene Database(Genecards),the two were intersected to obtain the drug-disease intersection target,and Venn diagram(Venn)was made;the PPI network of intersection target was constructed using STRING platform,the PPI network TSV file was imported into Cytoscape3.7.1 software,Cytoscape3.7.1 software was used for secondary construction according to the Degre value to make light-dark size changes and select the key core targets;GO and KEGG enrichment analysis were performed with the help of Metascape database to screen out the active ingredients of Qiyao Xiaoke Capsule in the treatment of type 2 diabetes.Results 251 potential targets of drugs and 1362 potential targets of diseases were obtained,140 intersection targets of the two,the core targets after screening:MAPK protein,interleukin 6(IL-6),AKT pathway,TNF tumor necrosis factor,etc.the biological processes involved included:reactive oxygen species metabolic process,oxygen level response,lipopolysaccharide response,apoptotic sig-naling pathway,nuclear receptor activity,heme binding,inflammatory response,etc.;the related pathways included:the role of AGE-RAGE signaling pathway in diabetic complications,the role of proteoglycans in cancer,NF-κB signaling pathway,sphingolipid signaling pathway,insulin resistance,estrogen signaling pathway,etc.Conclusion Through network pharmacological analysis,Qiyao Xiaoke Capsule can be expressed through multiple targets,multiple genes and multiple pathways in the treatment of type 2 diabetes.In addition to its hypoglycemic and lipid-lowering effects,it also has the effects of avoiding diabetic compli-cations,reducing liver and kidney and vascular endothelial damage,reducing insulin resistance and preventing cancer,providing a new direction of thinking for the use of traditional Chinese medicine combined with decoction formula in the future.
