摘要
本试验旨在研究饲粮中添加山香圆叶提取物(TFE)对文昌鸡生长性能、血清免疫和抗氧化功能及肠道微生物的影响。选择同批次的42日龄雌性文昌鸡432羽,随机分为4组,每组6个重复,每个重复18羽。对照组饲喂基础饲粮,试验组饲喂在基础饲粮中分别添加0.015%、0.030%、0.060%TFE的试验饲粮,试验期为42 d。结果表明:1)随着饲粮中TFE添加水平的增加,文昌鸡终末体重呈显著线性升高(P<0.05),料重比(F/G)呈显著线性和二次曲线变化(P<0.05)。与对照组相比,添加0.030%和0.060%的TFE显著提高了文昌鸡的平均日增重(ADG)(P<0.05)。2)随着TFE添加水平的增加,文昌鸡血清总蛋白(TP)、球蛋白(GLB)、白细胞介素-10(IL-10)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量呈显著线性和二次曲线变化(P<0.05),血清免疫球蛋白M(IgM)含量呈显著线性增加(P<0.05)。与对照组相比,添加0.030%和0.060%的TFE显著提高了血清免疫球蛋白A(IgA)和免疫球蛋白G(IgG)含量(P<0.05),显著降低了血清丙二醛(MDA)、IL-6和TNF-α含量;添加0.060%的TFE显著提高了血清总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)活性和IL-10含量(P<0.05),不同添加水平的TFE均显著提高了文昌鸡血清谷胱甘肽过氧化物酶(GSH-Px)活性(P<0.05),显著降低了血清IL-1β含量(P<0.05)。3)随着TFE添加水平的增加,空肠绒毛高度和绒毛高度/隐窝深度呈显著线性和二次曲线变化(P<0.05)。4)与对照组相比,添加0.060%的TFE显著降低了盲肠变形菌门相对丰度(P<0.05),显著提高了粪肠杆菌属和乳酸杆菌属的相对丰度(P<0.05)。5)通过二次函数拟合,确定TFE最适添加水平为0.056%。综上所述,饲粮中添加TFE能够改善文昌鸡生长性能,提高文昌鸡血清免疫和抗氧化功能,可以通过提高血清抗炎因子含量,降低促炎因子含量,从而缓解机体炎症损伤,改善肠道组织形态,促进肠道对营养物质的吸收。本试验条件下,推荐42~84日龄文昌鸡TFE最适添加水平为0.056%。
This experiment was conducted to investigate the effects of Turpiniae folium extract(TFE)on growth performance,serum immune and antioxidant function and intestinal microflora of Wenchang chickens.A total of 432 female Wenchang chickens at 42 days of age were randomly divided into 4 groups,with 6 replicates of 18 birds.Birds received either a basal diet(control group)or the basal diet+0.015%,0.030%or0.060%TFE.The experiment lasted for 42 days.The results showed as follows:1)the final body weight of chicken increased linearly(P<0.05),feed to gain ratio(F/G)changed linearly and quadratically(P<0.05)with increasing TFE levels.Compared with the control group,the average daily gain(ADG)of chicken significantly increased by adding 0.030%and 0.060%TFE(P<0.05).2)With increasing TFE levels,dietary TFE supplementation resulted in linear and quadratic variations in serum total protein(TP),globulin(GLB),interleukin-10(IL-10),interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)contents(P<0.05),and linear increases in serum immunoglobulin M(IgM)content(P<0.05).Compared with the control group,serum immunoglobulin A(IgA)and immunoglobulin G(IgG)contents increased significantly(P<0.05),and malondialdehyde(MDA),IL-6 and TNF-αcontents decreased significantly(P<0.05)in 0.030%and 0.060%TFE groups;the total antioxidant capacity(T-AOC),superoxide dismutase(SOD)activity and IL-10 content in serum showed significant increases in 0.060%TFE group(P<0.05);chicken fed TFE-supplemented diets exhibited both significant increases in glutathione peroxidase(GSH-Px)activity and decreases in serum IL-1βcontent(P<0.05).3)Jejunum villus height(VH)and villus height/crypt depth(VH/CD)differed linearly and quadratically(P<0.05)with increase levels of dietary TFE.4)Compared with the control group,the relative abundance of Proteobacteria in caecum decreased significantly(P<0.05),and the relative abundance of Faecalibacterium and Lactobacillus increased significantly(P<0.05)in 0.060%TFE group.5)Analysis by quadratic regression showed the optimal supplementation TFE level was 0.056%.In conclusion,TFE can improve the growth performance,serum immune and antioxidant function of Wenchang chickens.Increasing the content of serum anti-inflammatory factors and reducing the content of pro-inflammatory factors can alleviate the body’s inflammatory injury,improve the intestinal tissue morphology,and promote the intestinal absorption of nutrients.The optimal level of TFE supplementation for Wenchang chickens aged 42 to 84 d is 0.056%.
