摘要
目的 基于网络药理学研究金砂行气胶囊治疗慢性浅表性胃炎的药理作用与分子机制。方法 构建慢性浅表性胃炎大鼠模型,评价金砂行气胶囊治疗慢性浅表性胃炎的药理作用。借助TCMSP数据库筛选出复方金砂行气胶囊的主要有效成分及相关作用靶点,利用Cytoscape软件构建“成分-疾病-靶点”交互作用网络,借助David数据库对关键靶点进行GO和KEGG通路富集分析。结果 金砂行气胶囊能有效改善大鼠的慢性浅表性胃炎症状。网络药理学分析表明,金砂行气胶囊主要是通过槲皮素、山柰酚、木犀草素、熊果酸、β-谷甾醇、异鼠李素等15个活性化合物,调节癌症通路(pathways in cancer)、肿瘤坏死因子(TNF signal in pathway)、PI3K-Akt信号通路(PI3K-Akt signal in pathway)、MAPK信号通路(MAPK signal in pathway)等相关通路中的TP53、Akt1、IL-6、TNF、CASP3、MAPK1、MAPK3、JUN、IL-10等20个潜在靶点发挥药理作用的。结论 金砂行气胶囊能有效缓解慢性浅表性胃炎模型大鼠的症状,是通过多成分、多靶点、多途径协同发挥作用。
AIM To investigate the effects and mechanism of Jinsha Xingqi Capsules in the treatment of chronic superficial gastritis(CSG)based on network pharmacology.METHODS Upon the established CSG rat models,Jinsha Xingqi Capsules were administered and had their pharmacological effects assessed.Jinsha Xingqi Capsules and CSG had the main active components and their related targets retrieved with the help of TCMSP database respectively,their‘component-disease-target’interaction network constructed by using Cytoscape software;their key target genes GO and KEGG functional enrichment analysis performed by using DAVID database.RESULTS Jinsha Xingqi Capsules effectively improved the symptoms of CSG rats,shown by theresult of network pharmacology analysis,the 15 active constituents,including quercetin,kaempferol,luteolin,ursolic acid,isorhamnetin,β-sitosterol,etc.,which mainly act via pathway regulation of 20 potential targets of TP53,Akt1,IL-6,TNF,CASP3,MAPK1,MAPK3,JUN,IL-10 and so on associated with cancer,TNF,PI3 K-Akt,MAPK and etc.CONCLUSION The therapeutic effects of Jinsha Xingqi Capsules on CSG rats mainly contribute to the multi-component,multi-target,and multi-pathway synergistic pharmacological interaction.
