不同品种花生红衣中八种酚类物质成分分析 被引量:7

Analysis of eight phenolic contents in peanuts skin from different cultivars

摘要 为探讨不同品种花生红衣中酚类物质的含量差异,建立了HPLC同时测定花生红衣中8种酚类物质的方法,对40种花生红衣中的酚类物质进行了检测,并采用多种分析方法进行综合评价。结果表明,以X-Bridge shield RP18色谱柱(4.6 mm×250 mm,5μm)为固定相,以A[V(乙酸)∶V(乙腈)=1∶1000],B[V(乙酸)∶V(水溶液)=1∶1000]为流动相进行梯度洗脱,多波长检测,在此条件下8种酚类物质能较好分离,且重复性好(相对标准偏差≤3.92%),精密度高(相对标准偏差≤1.96%)。统计分析和热图分析表明,不同品种花生红衣中8种酚类物质存在较大差异;相关性分析表明,原儿茶酸、儿茶素、表儿茶素没食子酸酯与原花青素C_(1),儿茶素与咖啡酸之间均呈现极显著正相关,阿魏酸与原儿茶酸、原花青素C_(1)、表儿茶素没食子酸酯之间均呈现极显著负相关;40种花生红衣样品可聚类成三大类。因此,不同花生红衣中酚类物质的种类及含量存在较大差异,且酚类物质间具有一定的相关性。该研究可为挖掘我国花生种质资源优势提供理论参考。 To investigate the difference of phenolic contents in peanuts skin from different cultivars,a HPLC method was established for simultaneous determination of 8 phenolic compounds in 40 peanuts skin varieties.Various analysis methods were used for comprehensive evaluation.The chromatographic separation was achieved using an X-Bridge shield RP18(4.6 mm×250 mm,5μm)through gradient elution with a the mobile phasesconsisting of acetic acid in water(volume ratio 1∶1000,A)and acetic acid in acetonitrile(volume ratio 1∶1000,B).The detection wavelengthswere set at 250,280,320 and 370 nm.The results showed that 8 phenolic compounds could be separated well withgood repeatability(RSD≤3.92%)and stability(RSD≤1.96%).Statistical analysis and heat map analysis showed that there were obvious differences in 8 phenolic substances among the varieties.Correlation analysis showed that there was astrong positive correlation between protocatechuic acid,catechin,epicatechin gallate and procyanidin C_(1),catechin and caffeic acid.Moreover,there was astrong negative correlation between ferulic acidandthree components,including protocatechuic acid,procyanidin C_(1)and epicatechin gallate.Cluster analysisshowed that 40 peanut skins could be clustered into 3 categories.Therefore,there were obvious differences among the varieties.And there was a certain correlation between phenolic substances.This study could provide a theoretical reference for exploring the advantages of peanut germplasm resources in China.
