新型冠状病毒奥密克戎变异株核酸检测试剂盒的检测效果比较 被引量:1

Comparision of real-time RT-PCR kits for SARS-CoV-2 Omicron variant detection

摘要 目的评估三种商业化试剂盒对新型冠状病毒奥密克戎变异株核酸检测的效果。方法收集2021年12月北京市监测发现的经测序分析确定为新型冠状病毒奥密克戎变异株的咽拭子标本8份,同时收集北京市2021年11月监测发现的新型冠状病毒德尔塔变异株咽拭子标本5份,用三种针对新型冠状病毒奥密克戎变异株的商业化荧光定量RT-PCR试剂盒进行检测,对检测效果进行比较分析。取其中两份Ct值较低的样本10倍系列稀释5个梯度,用于比较检测试剂的灵敏性。结果三种试剂盒均能特异性地检出8份奥密克戎变异株和5份非奥密克戎株(德尔塔变异株)。试剂A和B的灵敏性高于试剂盒C一个稀释度。结论三种试剂盒均能特异性地检测奥密克戎变异株。试剂盒A核酸用量最少,虽然只检测一个变异位点(Q498R),但该位点是奥密克戎变异株特有的,从而使实验操作和结果判定都更简便。 Objective To analyze the detection efficiency of three commercially available real-time RT-PCR kits for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)Omicron variant.Methods Eight confirmed SARS-CoV-2 Omicron variant positive throat swab samples collected in December 2021 and 5 confirmed SARS-CoV-2 Delta variant positive throat swab samples collected in November 2021 from the surveillance of SARS-CoV-2 in Beijing were detected with three commercially available real-time RT-PCR kits for SARS-CoV-2 Omicron variant,and results were compared and analyzed.Comparison of detection sensitivity of three kits was carried out using 5 serial samples of 10-fold dilution of two Omicron-positive samples.Results Eight samples of Omicron variant and 5 samples of non-Omicron variant(Delta variant)were detected correctly by all three kits.The sensitivities of kit A and kit B were 10-fold better than kit C.Conclusions The three kits were capable of detecting the Omicron variant specifically.The volume of nucleic acid used by Kit A was the least.The kit A detected only one mutation site(Q498R)which was the specific site for the variant and made the operation and result interpretation simpler.
