热胁迫对中间球海胆抗氧化酶活性及线粒体结构与功能的影响 被引量:2

Effects of Thermal Stress on the Activities of Antioxidant Enzymes and Mitochondrial Structure and Function in Strongylocentrotus intermedius

摘要 【目的】探讨中间球海胆对高温的生理适应模式。【方法】将中间球海胆(Strongylocentrotus intermedius)分别置于不同温度梯度(15、20、25℃)24 h后,检测中间球海胆体腔细胞的线粒体结构、肠组织中线粒体的Ca^(2+)浓度、ATPase活性以及体腔液抗氧化酶活性的变化。【结果】1)温度升高至20℃,中间球海胆体腔细胞线粒体数量与对照组相比显著升高,大部分线粒体膜完整,部分有破损现象;嵴部分排列有序,部分脱落排列不清晰;线粒体基质密度相对比较大。温度升高至25℃,线粒体数量与20℃相比显著降低,少部分线粒体膜完整,大部分有破损现象;嵴小部分排列有序,大部分脱落排列不清晰;线粒体基质密度相对比较大。2)随着温度升高,肠组织线粒体中Ca^(2+)浓度显著下降(P <0.01);ATP浓度先下降后上升,浓度变化不显著(P> 0.05);Ca^(2+)-ATP和Ca^(2+)-Mg^(2+)-ATP的酶活性显著下降(P <0.05),而Na^(+)-K^(+)-ATP酶活性显著升高(P <0.01)。3)随着温度升高,体腔液中谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px)酶活性先下降后上升(P <0.01);过氧化氢酶(Catalase,CAT)酶活性显著下降(P <0.05);过氧化物酶(Peroxidase,POD)和超氧化物歧化酶(Superoxide Dismutase,SOD)的酶活性显著上升(P<0.05)。【结论】在高温胁迫下,中间球海胆的体腔细胞线粒体结构、肠组织线粒体ATP浓度、Ca^(2+)浓度、ATP酶活性和抗氧化酶活性均发生不同程度变化,说明高温影响了中间球海胆亚细胞水平的能量代谢以及生理生化指标(抗氧化酶活性)。 【Objective】 To explore the physiological adaptation mode of Strongylocentrotus intermediu to high temperature. 【Method】 Place S. intermediu in different temperature gradients(15°C, 20° C, 25° C) 24 h. Then detect the mitochondrial structure of coelomocytes and the Ca^(2+)of mitochondria in intestinal tissues, and changes in concentration, ATPase activity and antioxidant enzyme activity in coelomic fluid.【Result】1) When the temperature was increased to 20° C, the number of mitochondria in coelomocytes of S. intermedius increased significantly compared with the control group;most of the mitochondrial membranes were intact, and some were damaged. The cristae are partly arranged orderly, and some fall off and the arrangement is unclear. The mitochondrial matrix is relatively dense.The number of mitochondria decreased significantly at 25°C,compared with that at20° C, a few mitochondria membranes were intact, and most of them were damaged. The small parts of the cristae are arranged in an orderly manner, and most of the shedding arrangements are not clear;The mitochondrial matrix is relatively dense. 2) With the increase of temperature, the concentration of Ca^(2+)in the mitochondria of intestines decreased significantly(P < 0.01);The concentration of ATP first decreased and then increased, and the change in concentration was not significant(P > 0.05);The enzymatic activity of Ca^(2+)-ATP and Ca^(2+)-Mg^(2+)-ATP decreased significantly(P < 0.05), while the enzymatic activity of the Na^(+)-K^(+)-ATP increased significantly(P < 0.01). 3) As the temperature increases, the activity of glutathione peroxidase(GSH-Px) enzyme in coelom fluid first decreased and then increased(P < 0.01);the activity of catalase(CAT) enzyme decreased significantly(P < 0.05);the activities of peroxidase(POD) and superoxide Dismutase(SOD) increased significantly(P < 0.05).【Conclusion】 Under high-temperature stress, the mitochondrial structure of coelomocytes,mitochondrial ATP concentration, Ca^(2+)concentration, ATPase activity, and antioxidant enzyme activity in the intestines of S. intermedius changed to varying degrees, indicated that high temperature affected the energy metabolism and physiological and biochemical indicators(antioxidant enzyme activity) at the subcellular level of S. intermedius.
