种子不同贮藏温度对玉米幼苗生长及相关代谢基因表达的影响 被引量:2

摘要 目的:为明确不同贮藏温度对玉米种子活力以及幼苗相关基因的影响,选取不同温度贮藏的玉米种子,对种子的发芽能力以及幼苗相关基因进行分析。方法:将常温(25~30℃)和低温(0~4℃)贮藏的种子在人工气候箱内进行标准发芽试验,分别在实验后的第3天和第7天计算发芽势和发芽率并通过实验数据计算发芽指数和活力指数。在第7天后分别取常温和低温幼苗进行转录组测序,将筛选出的差异基因在相应的数据库中进行对比(KOG、GO、KEGG),分析处理差异基因的功能和参与的调控路径。结果:低温贮藏的玉米种子在发芽势、发芽率、发芽指数、活力指数上显著高于自然贮藏的对照种子。在转录组测序和数据分析中,低温贮藏的玉米种子与自然贮藏的玉米种子处理间筛选出4个差异基因,包含3个上调基因和1个下调基因。在GO数据分析中4个差异基因分别涉及到应激反应等生理或生化过程;在KEGG代谢通路中4个差异基因显著富集植物-病原互作、嘌呤代谢、N-聚糖生物合成代谢通路中。结论:在自然贮藏和低温贮藏条件下,植物-病原互作、N-聚糖生物合成、嘌呤代谢相关基因在玉米种子活力中发挥重要作用。 Objective:To determine the effects of different storage temperatures on maize seed vigor and seedling-related genes.Methods:Standard germination experiments were carried out in artificial climate box under 25~30℃and 0~4℃storage conditions.Germination potential and germination rate were calculated on the 3rd and 7th day after experiment respectively,and germination index and vigor index were calculated through experimental data.The transcriptome was sequenced after the 7th day at room temperature and low temperature,and the differential genes were compared in the corresponding databases,KOG,GO and KEGG.Results:The germination vigor,germination rate,germination index and vigor index of maize seeds stored at low temperature were significantly higher than those of control seeds stored at low temperature.In transcriptome sequencing and data analysis,four differential genes,including three up-regulated genes and one down-regulated gene,were screened between cold-stored and natural-stored maize seeds.In GO data analysis,four different genes were involved in physiological or biochemical processes such as anti-stimulation.In the KEGG metabolic pathway,four differential genes were significantly enriched in plant-pathogen interaction,purine metabolism,and N-glycan biosynthesis pathways.Conclusion:Plant-pathogen interaction,N-glycan biosynthesis and purine metabolism related genes play an important role in maize seed vigor under natural and low-temperature storage conditions.
