超高效液相色谱法同时测定补阳还五汤冻干物中10种成分的含量 被引量:5

Simultaneous Content Determination of 10 Components in Lyophilized Product of Buyang Huanwu Decoction on UPLC

摘要 目的建立同时测定补阳还五汤中川芎嗪、羟基红花黄色素A、芍药内酯苷、芍药苷、毛蕊异黄酮苷、阿魏酸、芒柄花苷、毛蕊异黄酮、苯甲酰芍药苷、芒柄花素、洋川芎内酯A等11种成分的超高效液相色谱(UPLC)分析方法,并测定其在冻干物中的含量。方法采用Phenomenex Luna C_(18)色谱柱(250 mm×4.6 mm,5μm);以0.1%甲酸水溶液-乙腈为流动相,梯度洗脱;流速:0.8 mL·min^(-1);柱温:30℃;检测波长:芍药内酯苷、芍药苷、苯甲酰芍药苷为240 nm,川芎嗪、羟基红花黄色素A、毛蕊异黄酮苷、阿魏酸、芒柄花苷、毛蕊异黄酮、芒柄花素、洋川芎内酯A为300 nm;进样量:10μL;测定6批补阳还五汤冻干物中各成分含量。结果建立了UPLC法同时测定补阳还五汤中11种成分含量的方法,精密度、稳定性和准确度良好。除川芎嗪未在冻干物中被检出外,羟基红花黄色素A、芍药内酯苷、芍药苷、毛蕊异黄酮苷、阿魏酸、芒柄花苷、毛蕊异黄酮、苯甲酰芍药苷、芒柄花素、洋川芎内酯A等10种成分在6批补阳还五汤冻干物中的平均含量分别为68.57、29.15、2007.44、327.9、63.12、102.42、31.06、18.40、15.51、31.53μg·g^(-1)。结论所建立的分析方法灵敏、可靠、稳定、重现性好,实现了补阳还五汤冻干物多成分质量控制,可为同类研究提供借鉴。 Objective To establish a UPLC method for simultaneous determination of eleven components including ligustrazine,hydroxyl-safflor yellow A,albiflorin,paeoniflorin,calycosin-O-β-D-glucopyranoside,frulic acid,ononin,calycosin,benzoylpaenoniflorin,formononetin,senkyunolide A in Buyang Huanwu Decoction,and their contents in lyophilized product were determined.Methods Chromatographic separation was performed on Phenomenex Luna C_(18) chromatographic column(250 mm×4.6 mm,5μm)at 30℃of column temperature with 0.1%aqueous formic acid-acetonitrile as mobile phase for gradient elution.The flow rate was set at 0.8 mL·min^(-1).The detection wavelengths were set at 240 nm for albiflorin,paeoniflorin,benzoylpaenoniflorin,and 300 nm for ligustrazine,hydroxyl-safflor yellow A,calycosin-O-β-D-glucopyranoside,ferulic acid,ononin,calycosin,formononetin,senkyunolide A.The injection volume was 10μL.The contents of each component in 6 batches of lyophilized product of Buyang Huanwu Decoction was determined.Results Simultaneous detection methods of 11 components were established,with good precision,stability and accuracy.In addition to ligustrazine not detected in the lyophilized product,the average contents of 10 components in 6 batches of lyophilized product of Buyang Huanwu Decoction such as hydroxyl-safflor yellow A,albiflorin,paeoniflorin,calycosin-O-β-D-glucopyranoside,ferulic acid,ononin,calycosin,benzoylpaenoniflorin,formononetin,senkyunolide A were 68.57,29.15,2007.44,327.9,63.12,102.42,31.06,18.40,15.51,31.53μg·g^(-1)respectively.Conclusion The established analytical method is sensitive,reliable,stable,and reproducible.It realizes the multi-component quality control of lyophilized product of Buyang Huanwu Decoction and provides reference for similar studies.
