负载血管内皮生长因子的邻苯二酚壳聚糖体外药物缓释性能及抗菌性研究 被引量:2

摘要 目的 研究负载血管内皮生长因子(VEGF)的邻苯二酚壳聚糖水凝胶的体外药物缓释能力以及抗菌性。方法通过偶联反应和化学交联法合成载有VEGF的邻苯二酚壳聚糖水凝胶,采用傅里叶变换红外光谱分析邻苯二酚接枝情况,流变分析和接触角表征其弹性性能和亲水性,采用酶联免疫吸附试验检测VEGF体外释放情况。通过菌落计数法和抑菌环法检测其对大肠杆菌(E.coli)和具核梭杆菌(F.n)的体外抗菌性。结果 红外光谱图显示邻苯二酚壳聚糖改性成功,邻苯二酚壳聚糖水凝胶的弹性性能和亲水性相较于壳聚糖水凝胶均有增强,并且对VEGF具有良好的控释性能,对E.coli和F.n具有良好的抑菌作用。结论 负载VEGF的邻苯二酚壳聚糖水凝胶具有良好的体外药物缓释能力以及抗菌性,同时具备促进血管生成和控制感染两种潜能,为牙髓组织工程再生中支架材料的研究提供新思路。 Objective To study the drug release ability and in vitro antibacterial activity of catechol chitosan hydrogel loaded with vascular endothelial growth factor(VEGF).Methods Catechol chitosan hydrogel containing VEGF was synthesized by coupling method and cross-linking method.The grafting of catechol was analyzed by Fourier transform infrared spectroscopy.The elastic properties and hydrophilicity were characterized by rheological analysis and contact Angle.Enzyme linked immunosorbent assay(ELISA)was used to detect the release of VEGF in vitro.Then in vitro antibacterial activity of Escherichia coli(E.coli)and Clostridium nucleorum(F.n)was studied by colony counting and bacteriostatic ring method.Results IR spectra showed that catechol chitosan modified successfully.Hydrophilicity and mechanical properties of catechol chitosan hydrogel were enhanced compared with chitosan hydrogel.Moreover,the catechol chitosan hydrogel had good controlled release performance of VEGF.In addition,catechol chitosan hydrogel had remarkable antibacterial activity against E.coli and F.n.Conclusion The catechol chitosan hydrogel loaded with VEGF has the ability of drug sustained release and good antibacterial activity in vitro,as well as the potential of promoting angiogenesis and controlling infection,which can provide a new idea for the research of scaffold materials for dental pulp tissue engineering.
