电针井穴对VD大鼠海马CA1区PI3K/Akt/mTOR通路及VEGF、bFGF的影响 被引量:10

Effects of electroacupuncture at Jing point on PI3K/Akt/mTOR pathway,VEGF and bFGF in hippocampal CA1 region of VD rats

摘要 目的探讨电针井穴改善VD大鼠记忆及对VD大鼠海马CA1区PI3K/Akt/mTOR信号通路及对细胞生长因子VEGF、bFGF表达的调节作用。方法用4-VO(四血管阻断法)制备VD大鼠模型,将大鼠随机分为电针组(VD+Z组)、西药组(VD+Y组)、VD对照组(VD组)、假手术组(BG组)。以水迷宫试验、神经行为学评分对全部治疗后大鼠的学习记忆能力进行评分;采用免疫组化、Western blot检测各组海马CA1区PI3K、Akt、mTOR、VEGF、bFGF的表达情况。结果与VD组相比,VD+Z组、VD+Y组的水迷宫实验逃避潜伏期、跨越平台、神经行为学评分显著好转(P<0.01);VD+Z组、VD+Y组CA1区PI3K、Akt、mTOR、VEGF、bFGF的各项表达均显著升高(P<0.01)。结论电针相关井穴可激活海马CA1区PI3K/Akt/mTOR信号通路及VEGF、bFGF表达,促进血管再生,进而有效提高学习记忆功能以改善VD。 Objective To investigate the effects of electroacupuncture at Jing point on memory,PI3 K/Akt/mTOR signal pathway and the expression of VEGF and bFGF in hippocampal CA1 region of VD rats.Methods VD rat model was prepared by 4-VO(four vessel occlusion method).The rats were randomly divided into electroacupuncture group(VD+Z group),western medicine group(VD+Y group),VD control group(VD group)and sham operation group(BG Group).The learning and memory abilities of all treated rats were evaluated by Morris water maze behavior detection and neurobehavioral score;Immunohistochemistry and Western blot were used to detect the expression of PI3 K,Akt,mTOR,VEGF and bFGF in hippocampal CA1 region of each group.Results compared with VD group,the escape latency,crossing platform and neurobehavioral score of VD+Z group and VD+Y group were significantly improved(P<0.01);The expressions of PI3 K,Akt,mTOR,VEGF and bFGF in CA1 were significantly increased in VD+Z group and VD+Y group(P<0.01).Conclusion Electroacupuncture at Jing points can activate PI3 K/Akt/mTOR signal pathway and the expression of VEGF and bFGF in hippocampal CA1 area,promote vascular regeneration,and effectively improve learning and memory function to improve VD.
