大黄素对慢性肾衰犬氧化应激及微炎症状态的影响 被引量:2

Effect of Emodin on Oxidative Stress and Micro-Inflammation in Dogs with Chronic Renal Failure

摘要 本试验旨在研究大黄素对慢性肾衰竭(CRF)宠物犬氧化应激和微炎症状态的影响,选取47例犬慢性肾衰竭临床病例,随机分为常规治疗组(UT组,n=8)、低剂量大黄素治疗组(LRT组,n=12)、中剂量大黄素治疗组(MRT组,n=12)和高剂量大黄素治疗组(HRT组,n=15),同时选择10例健康犬作为对照组(CG组)。UT组患犬治疗方案为口服复方α-酮酸片(1片/次,2次/d)+自由采食肾脏处方粮,LRT组、MRT组和HRT组分别在UT组治疗基础上按25、50 mg/(kg·bw·d)和100 mg/(kg·bw·d)口服大黄素。试验前后分别采集血液、尿液,测定肾功能指标、氧化应激指标和炎症指标。结果显示:各治疗组的血清肌酐(Scr)、尿素氮(BUN)和尿肾脏损伤因子-1(KIM-1)均显著低于治疗前(P<0.05);治疗后,HRT组Scr、BUN和KIM-1均低于MRT组、LRT组和UT组(P<0.05);与UT组比较,HRT组丙二醛(MDA)、尿8-羟基脱氧鸟苷(8-OH-dG)、C反应蛋白(CRP)、白细胞介素6(IL-6)和肿瘤坏死因子α(TNF-α)均显著降低(P<0.05),超氧化物歧化酶(SOD)、总抗氧化能力(T-AOC)显著升高(P<0.05);Scr、BUN和KIM-1等肾功能指标与MDA、8-OH-dG、CRP、IL-6和TNF-α呈正相关关系,与SOD和T-AOC呈负相关关系。结果表明,大黄素有助于改善CRF犬肾功能,可能与大黄素抑制机体氧化应激和炎症反应有关,以100 mg/(kg·bw·d)给药效果最佳。 This study aimed to investigate the effect of emodin on oxidative stress and micro-inflammation in dogs with chronic renal failure(CRF).A total of 47 dogs with CRF were randomly divided into four groups:usual treatment group(UT group,n=8),low dose emodin treatment group(LRT group,n=12),medium dose emodin treatment group(MRT group,n=12) and high dose emodin treatment group(HRT group,n=15).In addition,10 healthy dogs were selected as control group(CG group).The treatment plan of dogs in UT group was to take compound α-ketoacid tablets(1 tablet/time,2 times/d)+kidney prescription food freely.The dogs in LRT group,MRT group and HRT group were to take emodin orally according to 25,50 mg/(kg·bw·d),and 100 mg/(kg·bw·d) on the basis of UT group,respectively.Before and after the experiment,blood and urine were collected to measure renal function index,oxidative stress index and inflammation index.The results show that:the levels of serum creatinine(Scr),urea nitrogen(BUN) and urinary kidney injury factor-1(KIM-1) were significantly lower in each treatment group than those before the treatment(P<0.05);after treatment,the levels of Scr,BUN and KIM-1 in HRT group were lower than those in MRT group,LRT group and UT group(P<0.05);compared with UT group,the levels of MDA,8-OH-dG,CRP,IL-6 and TNF-α decreased significantly(P<0.05) in HRT group,while the levels of SOD and T-AOC increased significantly(P<0.05);the levels of Scr,BUN and KIM-1 were positively correlated with MDA,8-OH-dG,CRP,IL-6 and TNF-α,and negatively correlated with SOD and T-AOC.Thus,emodin has therapeutic effect on CRF in dogs,which may be related to the inhibition of oxidative stress and inflammatory response,the dose of 100 mg/(kg·bw·d) appears to be optimal.
