皂素与十二烷基硫酸钠对阳性血培养瓶直接质谱鉴定处理效果的影响

Influences of Saponin and Sodium Dodecyl Sulfate on the Direct Mass Spectrometry Identification of Positive Blood Culture Flasks

摘要 目的通过分析皂素、十二烷基硫酸钠(SDS)及其不同配比的使用对阳性血培养瓶直接质谱鉴定处理效果的影响,筛选更适用于阳性血培养瓶直接质谱鉴定的表面活性剂使用方案。方法通过模拟血培养瓶的方法验证不同浓度皂素溶液及SDS溶液对阳性血培养液的鉴定效果,并与传统鉴定方法的处理效果作对比,分析两种表面活性剂对阳性血培养瓶直接质谱鉴定的处理效果及影响。结果实验结果表明,优化后4%皂素溶液的鉴定准确率为89.3%(50/56),3%SDS溶液的鉴定准确率为76.8%(43/56),传统鉴定方法的鉴定准确率为92.9%(52/56),4%皂素溶液与传统鉴定方法的鉴定准确率均高于3%SDS溶液,4%皂素溶液与传统鉴定方法的鉴定准确率比较无显著差异。结论作为直接鉴定阳性血培养液的表面活性剂,皂素溶液的鉴定准确率优于SDS溶液,表明皂素在后期开发应用于阳性血培养瓶直接质谱鉴定方面具有较大的潜力。 Objective By analyzing the influences of saponin,sodium dodecyl sulfate(SDS)and their different ratios on the direct mass spectrometry identification of positive blood culture flasks,a more suitable method of using surfactants was selected.Methods The identification effect of different concentrations of saponin solution and SDS solution on positive blood culture medium was verified by the method of simulated blood culture flask,and compared with the treatment effect of traditional identification method,and the treatment effect and influences of two kinds of surfactants on the direct mass spectrometry identification of positive blood culture flasks were analyzed.Results The experimental results showed that the identification accuracy of the optimized 4% saponin solution was 89.3%(50/56),the identification accuracy of the 3% SDS solution was 76.8%(43/56),and the identification accuracy of the traditional identification method was 92.9%(52/56).The accuracy of 4% saponin solution and the traditional identification method were higher than that of 3% SDS solution,and there was no significant difference when the identification accuracy of 4% saponin solution and traditional identification method were compared.Conclusion As a surfactant for direct identification of positive blood culture medium,the identification accuracy of saponin solution is better than that of SDS solution,indicating that saponin has great potential in the later development of direct mass spectrometry identification of positive blood culture flasks.
