金线莲苷对TNF-α诱导的人类风湿关节炎滑膜细胞增殖及炎症因子、MMPs产生的影响 被引量:7

Effects of kinsenoside on TNF-αinduced proliferation of human rheumatoid arthritis synoviocytes and production of inflammatory factors and MMPs

摘要 目的探讨金线莲苷(Kin)对肿瘤坏死因子-α(TNF-α)诱导的人类风湿关节炎滑膜细胞的增殖及炎症因子、基质金属蛋白酶MMPs产生的影响。方法以TNF-α诱导人类风湿关节炎滑膜成纤维细胞,同时给予不同剂量的金线莲苷刺激,CCK-8法用来检测滑膜细胞的增殖,ELISA用来检测细胞上清液中炎症因子白介素-6(IL-6)、白介素-1β(IL-1β)、白介素-8(IL-8)的分泌,实时荧光定量PCR和Western Blot分别用来检测细胞中基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶-3(MMP-3)、基质金属蛋白酶-13(MMP-13)的m RNA水平和蛋白水平表达。结果5μg/mL的金线莲苷能显著抑制TNF-α诱导的滑膜细胞的增殖(P<0.05)和滑膜细胞上清液中炎症因子IL-6、IL-1β、IL-8的升高(P<0.05)以及细胞中基质金属蛋白酶MMP-3、MMP-13的异常表达(P<0.05),并极显著地抑制MMP1的异常表达(P<0.01);10μg/mL的金线莲苷能够极显著抑制TNF-α诱导的滑膜细胞的增殖(P<0.01)和细胞上清液中IL-6、IL-1β的升高(P<0.01)以及细胞中MMP-1、MMP-3、MMP-13的异常表达(P<0.05),并显著降低IL-8的升高。结论本研究揭示了金线莲苷对人类风湿关节炎滑膜细胞的作用机制,为金线莲苷治疗类风湿关节炎提供了一定参考。 Objective To investigate the effect of kinsenoside(Kin)on tumor necrosis factor-α(TNF-α)induced proliferation of human rheumatoid arthritis synoviocytes and the production of inflammatory factors and MMPs.Methods TNF-αwas used to induce human rheumatoid arthritis(RA)synovial fibroblasts,and different doses of Kin were given at the same time.CCK-8 method was used to detect the proliferation of synovial cells.ELISA was used to detect the secretion of inflammatory factors interleukin-6(IL-6),IL-1β,IL-8 in the supernatant of cells.RT-PCR and WB was used to detect the expression of Metalloproteinase-1(MMP-1),MMP-3,MMP-13 in cells.Results The results showed that 5μg/mL of Kin could significantly inhibit the proliferation of TNF-αinduced synovial cells(P<0.05),the increase of IL-6,IL-1β,IL-8 in the supernatant of synovial cells(P<0.05),and the abnormal expression of MMP-3 and MMP-13 in cells(P<0.05),extremely significantly inhibit and the abnormal expression of MMP-1(P<0.01);10μg/mL of Kin could extremely significantly inhibit TNF-αinduced proliferation of synovial cells(P<0.01),the increase of inflammatory factors IL-6 and IL-1βin supernatant(P<0.01),and the abnormal expression of MMP-1,MMP-3 and MMP-13 in synovial cells(P<0.05),and significantly inhibit the increase of IL-8.Conclusion This study reveales the mechanism of action of Kin on human rheumatoid arthritis synoviocytes,and provides some reference for the treatment of RA with Kin.
