摘要
目的探讨磷酸酶及张力蛋白同源基因蛋白(PTEN)、补丁蛋白(Ptch)在口腔鳞状细胞癌中的表达及意义。方法选取口腔鳞状细胞癌组织标本78例,同时选取正常口腔组织标本56例作为对照,采用免疫组化染色法检测PTEN、Ptch蛋白表达。结果口腔鳞状细胞癌中PETN阳性表达率为34.62%,明显低于正常口腔组织(P<0.05),而Ptch阳性表达率为67.95%,明显高于正常口腔组织(P<0.05);TNM分期Ⅲ~Ⅳ期、中低分化和有淋巴结转移口腔鳞状细胞癌组织PETN阳性表达率分别为13.04%、10.00%和11.43%,明显低于Ⅰ~Ⅱ期、高分化和无淋巴结转移口腔鳞状细胞癌组织(P<0.05);TNM分期Ⅲ~Ⅳ期、有淋巴结转移口腔鳞状细胞癌组织Ptch阳性表达率为86.96%和88.57%,明显高于Ⅰ~Ⅱ期、无淋巴结转移口腔鳞状细胞癌组织(P<0.05);口腔鳞状细胞癌组织中PETN表达与Ptch表达呈负相关(γ_(s)=-0.655,P<0.05)。结论口腔鳞状细胞癌组织中PTEN表达下调,而Ptch表达上调,两者与临床病理特征有一定关系,且两者间存在负相关性。
Objective To investigate the expression and significance of phosphatase and tensin homologous gene protein(PTEN)and patch protein(Ptch)in oral squamous cell carcinoma.Methods 78 specimens of oral squamous cell carcinoma were selected,and 56 specimens of normal oral tissues were selected as controls,the expression of PTEN and Ptch protein was detected by immunohistochemical staining.Results The positive expression rate of PETN in oral squamous cell carcinoma was 34.62%,significantly lower than that in normal oral tissue(P<0.05),while the positive expression rate of Ptch was 67.95%,significantly higher than that in normal oral tissue(P<0.05);The positive expression rates of PETN in TNM stageⅢ~Ⅳ,moderately and poorly differentiated and lymph node metastasis oral squamous cell carcinoma tissues were 13.04%,10.00%and 11.43%,respectively,which were significantly lower than those in stageⅠ~Ⅱ,highly differentiated and non-lymph node metastasis oral squamous cell carcinoma tissues(P<0.05);The positive expression rate of Ptch in TNM stageⅢ~Ⅳand lymph node metastasis oral squamous cell carcinoma were 86.96%and 88.57%,which was significantly higher than that in stageⅠ~Ⅱ,non-lymph node metastasis oral squamous cell carcinoma(P<0.05);There was a negative correlation between the expression of PETN and Ptch in oral squamous cell carcinoma(γ_(s)=-0.655,P<0.05).Conclusion The expression of PTEN is down-regulated and Ptch is up-regulated in oral squamous cell carcinoma,which are related to clinicopathological features,there are a negative correlation between PTEN and Ptch.
