HIV-1感染者病毒学指标与血浆中CXC趋化因子水平的相关性

Correlation Between Virological Indexes and Plasma CXC Chemokine Levels in HIV-1 Infected Patients

摘要 目的探讨未治疗的HIV-1感染者不同HIV-1 RNA或HIV-1 DNA水平组间,患者血浆中趋化因子CXCL8、CXCL10、CXCL11和CXCL13表达水平的差异及其与HIV-1 RNA或HIV-1 DNA的相关性。方法未治疗HIV-1感染者66例。采用实时荧光定量聚合酶链反应检测血浆中HIV-1 RNA及外周血单个核细胞(PBMC)中HIV-1 DNA;用Luminex液相芯片检测血浆中趋化因子CXCL8、CXCL10、CXCL11和CXCL13表达水平;利用流式细胞术进行外周血CD4^(+)T细胞计数、CD8^(+)T细胞计数。采用SPSS23.0进行非参数检验及相关性分析等统计学分析。结果HIV-1 RNA或HIV-1 DNA高低分组患者的一般资料及免疫学指标间差异无统计学意义(P>0.05);趋化因子CXCL10、CXCL11和CXCL13在高HIV-1 RNA水平组及高HIV-1 DNA水平组中表达水平较高,且与对应低病毒学指标组患者差异有统计学意义(P<0.05);趋化因子CXCL8表达水平则在组间差异无统计学意义(P>0.05)。以200 cells/μL作为CD4^(+)T细胞计数截断值进行分层分析,得到相似的组间差异结果。CXCL10、CXCL11和CXCL13的表达水平与HIV-1 RNA、HIV-1 DNA均存在正相关性(P<0.05),而CXCL8则不存在这种相关性(P>0.05)。结论趋化因子CXCL10、CXCL11和CXCL13在高HIV-1 RNA、HIV-1 DNA的感染者体内表达水平较高,可能参与并促进了HIV-1在患者体内的复制及储存库的建立。 Objective To investigate the differences in expression levels of plasma chemokine CXCL8,CXCL10,CXCL11 and CXCL13 and their correlation with human immunodeficiency virus(HIV)-1 RNA and HIV-1 DNA in untreated HIV-1 infected patients with different HIV-1 RNA or HIV-1 DNA levels.Methods A total of 66 untreated HIV-1 infected patients were recruited.HIV-1 RNA in plasma and HIV-1 DNA in PBMC were detected by Real-time fluorescence quantitative polymerase chain reaction(RT-PCR).The expression of chemokine CXCL8,CXCL10,CXCL11 and CXCL13 in plasma was detected by Luminex liquid chip technology.Peripheral blood CD4^(+)T cell count and CD8^(+)T cell count were analyzed by flow cytometry.SPSS 23.0 was used for nonparametric test and correlation analysis.Results There was no significant difference in the general data and immunological indexes between the high and low HIV-1 RNA or HIV-1 DNA groups(P>0.05).The expression levels of CXCL10,CXCL11 and CXCL13 were higher in the high HIV-1 RNA level group and the high HIV-1 DNA level group,and were significantly different from those in the corresponding low virological index groups(P<0.05).There was no significant difference in the expression level of chemokine CXCL8 between the two groups(P>0.05).The stratified analysis using 200 cells/μL as the cut-off value of CD4^(+)T cell count showed similar intergroup differences.The expression levels of CXCL10,CXCL11 and CXCL13 were positively correlated with HIV-1 RNA and HIV-1 DNA(P<0.05),but CXCL8 were not(P>0.05).Conclusion Chemokine CXCL10,CXCL11 and CXCL13 are highly expressed in patients with high HIV-1 RNA and HIV-1 DNA,which may be involved in and promote the replication of HIV-1 in patients and the establishment of virus reservoirs.
