摘要
针对乙醇发酵醪,采用无有机溶剂流动相,建立了高效液相色谱法测定乙醇含量的方法。样品只需要经过简单的稀释离心即可测定,其色谱条件为:色谱柱为ShodexRSpak KC-811色谱柱;流动相为5 mmol/L硫酸水溶液;流速为1.0 mL/min;进样量为10μL;柱温为30℃;检测器为示差检测器,检测器温度为35℃。结果表明:该分析方法在乙醇浓度0.6~6.0 g/L范围内线性度良好,R^(2)=1;检出限为11.67 mg/L;定量限为40.85 mg/L;加标回收率为96.66%,重复性(RSD)小于1.0%。该分析方法样品处理简单,乙醇含量测定绿色、快速、准确、可靠,可以推广使用。
In the paper,a method for the determination of ethanol content in ethanol fermentation mash by HPLC was established,based on organic solvent-free mobile phase.The sample only needed to be diluted and centrifuged simply,and then it could be determined.The chromatographic conditions were as follows:the column was ShodexRSpak KC-811 column;the mobile phase was 5 mmol/L sulfuric acid aqueous solution;the flow rate was 1.0 ml/min;the injection volume was 10μL;the column temperature was 30℃;the detector was a differential detector,and the detector temperature was 35℃.The results shows that the analytical method has good linearity in the ethanol concentration range of 0.6g/L to 6.0g/L,R^(2)=1;the detection limit is 11.67mg/L;the limit of quantitation is 40.85mg/L;the recovery rate of the spiked standard is 96.99%,and the repeatability(RSD)is less than 1.0%.The analytical method is simple in sample treatment,and the determination of ethanol content is green,rapid,accurate and reliable.
