摘要
目的探讨栎树桑黄提取物(OPL)对结肠癌细胞SW620增殖、凋亡的影响及其对PI3K/AKT信号通路的调控作用。方法采用不同浓度的OPL处理人结肠癌细胞SW620,分别记为60μg/mL OPL组、120μg/mL OPL组、240μg/mL OPL组、480μg/mL OPL组;添加PI3K/AKT信号通路激动剂740Y-P与OPL共同处理SW620细胞,记为740Y-P+480μg/mL OPL组;采用MTT实验检测细胞活力;采用平板克隆形成实验检测细胞克隆形成能力;采用流式细胞术检测细胞凋亡率;采用Western blot法检测CyclinD1、Cleaved-caspase-3、p-PI3K、p-AKT蛋白表达量。结果与0μg/mL OPL组比较,60μg/mL OPL组、120μg/mL OPL组、240μg/mL OPL组、480μg/mL OPL组细胞活力降低(P均<0.05),CyclinD1、p-PI3K、p-AKT蛋白水平降低(P均<0.05),克隆形成数减少(P均<0.05),细胞凋亡率升高(P均<0.05),Cleaved-caspase-3蛋白水平升高(P均<0.05);与480μg/mL OPL组比较,740Y-P+480μg/mL OPL组细胞活力升高(P<0.05),克隆形成数增多(P<0.05),细胞凋亡率降低(P<0.05),CyclinD1、p-PI3K、p-AKT蛋白水平升高(P均<0.05),Cleaved-caspase-3蛋白水平降低(P<0.05)。结论OPL可抑制结肠癌细胞增殖、克隆形成及促进细胞凋亡,其作用机制与抑制PI3K/AKT信号通路的活化有关。
Objective To explore the effects of oak Phellinus linteus(OPL)on the proliferation and apoptosis of colon cancer cell line SW620 and its regulation of PI3K/AKT signaling pathway.Methods Different concentrations of OPL were used to treat human colon cancer cells SW620,which were recorded as 60μg/ml OPL group,120μg/ml OPL group,240μg/ml OPL group,and 480μg/ml OPL group.The PI3K/AKT signaling pathway agonist 740Y-P and OPL were added to treat SW620 cells together,which was recorded as the 740Y-P+480μg/ml OPL group.MTT assay was used to detect cell viability.The plate clone formation test was used to detect the cell clone formation ability.Flow cytometry was used to detect the apoptosis rate.Western blot was used to detect the protein expression of CyclinD1,Cleaved-caspase-3,p-PI3K,and p-AKT.Results Compared with the 0μg/ml OPL group,the cell viability of the 60μg/ml OPL group,120μg/ml OPL group,240μg/ml OPL group,and 480μg/ml OPL group was decreased(P<0.05),the protein levels of CyclinD1,p-PI3K,p-AKT were decreased(P<0.05),the number of clone formation was decreased(P<0.05),the rate of apoptosis was increased(P<0.05),and the protein level of Cleaved-caspase-3 was increased(P<0.05).Compared with the 480μg/ml OPL group,the cell viability of the 740Y-P+480μg/ml OPL group was increased(P<0.05),the number of clones was increased(P<0.05),and the rate of apoptosis was decreased(P<0.05),the protein levels of CyclinD1,p-PI3K,p-AKT were increased(P<0.05),and the protein level of Cleaved-caspase-3 was decreased(P<0.05).Conclusion Oak Phellinus linteus could inhibit the proliferation and clone formation of colon cancer cells,promote cell apoptosis,and its mechanism is related to inhibiting the activation of PI3K/Akt signaling pathway.
