摘要
目的:建立苗药观音座莲中紫萁内酯苷的HPLC含量测定方法,并分析叶子与根中紫萁内酯苷的含量差异。方法:采用ZORBAX SB-C;柱(5μm,4.6mm×250mm)测定紫萁内酯苷的含量,以0.1%磷酸水溶液(A)-甲醇(B)为流动相进行等度洗脱,流速1.0mL/min,检测波长220nm,柱温40℃;采用SPSS 26.0统计软件对含量测定结果进行分析。结果:上述色谱条件下,紫萁内酯苷在0.101~0.505mg/mL(r=0.9999)范围内呈现良好的线性关系,平均加样回收率为97.33%,RSD值为2.02%(n=6);通过统计分析发现,叶子与根中的紫萁内酯苷含量存在显著性差异(1.25%VS3.97%)。结论:建立的测定方法准确可靠、重复性好,可用于测定观音座莲中紫萁内酯苷的含量。根与叶中紫萁内酯苷的含量差异对比分析为观音座莲以根入药提供了理论依据。
Objective:To establish a HPLC method for the content determination of angiopteroside in Angiopteris fokiensis Hieron,and to analyze the content differences of angiopteroside between leaves and roots.Methods:A HPLC method was established based on the ZORBAX SB-C;column(5μm,4.6mm×250mm)with the mobile phase as a mixture of 0.1%phosphoric acid-methanol(98∶2).The flow rate was 1.0mL/min and the detection wave length at220nm as well as column temperature at 40℃.A SPSS statistical software was employed to analyze the results of content determination.Results:The calibration curve was linear within the range of 0.101~0.505mg/mL of angiopteroside(r=0.999 9),the average recovery rate was 97.33%with the RSDvalue as 2.02%(n=6).The content of angiopteroside differed significantly between the leave and root part of Angiopteris fokiensis Hieron.Conclusion:The established HPLC method was precise and reliable with good repeatability,which could be applied for the quality control of Angiopteris fokiensis Hieron.The content differences of angiopteroside in leaves and roots also provide the basis for the root of Angiopteris fokiensis as its medicinal part.
