摘要
目的比较正常小鼠与肾虚体质小鼠血液IgG水平及生化指标的变化,探索亲代体质对子代体质的形成及相关疾病发生发展的影响。方法根据中医学“恐伤肾”的原理,使用超声波驱鼠仪刺激孕鼠,致使其惊恐伤肾,直到小鼠出生后停止恐吓,使小鼠先天不足,随机抽取10只小鼠作为肾虚体质组,雌雄各半,观察各组小鼠一般状况。造模结束后称重,小鼠眼底采血,Eppen-dorf管收集,离心取血清进行生化指标及免疫球蛋白IgG的检测。结果①肾虚体质组小鼠体重明显轻于正常对照组小鼠。且身长相对较短,毛发稀疏,胆小易惊,呈蜷缩状,活动量较少,食量较小,与肾虚体质判断依据相符。具有统计学意义(P<0.05)。②小鼠血液生化指标检测发现,与正常对照组比较,肾虚体质组血液ALP(碱性磷酸酶)、Ca(钙)、TG(甘油三酯)增高,差异具有统计学意义(P<0.05或P<0.01)。肌酐及尿酸在正常对照组及肾虚体质组上差异虽然无统计学意义,但是肾虚体质组小鼠血液肌酐及尿酸数值呈增高趋势。③小鼠血液IgG浓度检测发现,与正常对照组相比,肾虚体质组小鼠血液免疫球蛋白IgG浓度低于正常对照组,差异具有统计学意义(P<0.05)。结论肾虚体质小鼠血液生化指标及免疫球蛋白IgG浓度均发生了变化,说明肾虚体质母鼠对子代鼠肾虚体质的形成产生影响并影响子代鼠的免疫球蛋白水平。
Objective To compare the changes of blood IgG level and biochemical indexes between normal mice and mice with kidney deficiency constitution,and to explore the effect of parental constitution on the formation of offspring constitution and the occurrence and development of related diseases.Methods According to the principle of“fear hurting kidney”in traditional Chinese medicine,the pregnant mice were stimulated by ultrasonic rat driving instrument,causing them to panic and injure the kidney until the mice stopped threatening after birth,making the mice congenital deficiency.10 mice were randomly selected as the kidney deficiency constitution group,half male and half female,and the general conditions of mice in each group were observed.After modeling,the mice were weighed,the blood was collected from the fundus of the eyes,collected with Eppen-dorf tube,and the serum was centrifuged for the detection of biochemical indexes and immunoglobulin IgG.Results①The weight of mice in kidney deficiency constitution group was significantly lighter than that in normal control group.And the body is relatively short,the hair is sparse,timid and easily startled,curled up,with less activity and less appetite,which is consistent with the judgment basis of kidney deficiency constitution.It was statistically significant(P<0.05).②The detection of blood biochemical indexes in mice showed that compared with the normal control group,the blood ALP(alkaline phosphatase),Ca(calcium)and TG(triglyceride)in the kidney deficiency group increased,and the difference was statistically significant(P<0.05 or P<0.01).Although there was no significant difference in creatinine and uric acid between normal control group and kidney deficiency constitution group,the values of blood creatinine and uric acid in kidney deficiency constitution group increased.③Compared with the normal control group,the detection of blood IgG concentration in mice showed that,the blood immunoglobulin IgG concentration in the kidney deficiency constitution group was lower than that in the normal control group,and the difference was statistically significant(P<0.05).Conclusion The blood biochemical indexes and the concentration of immunoglobulin IgG in mice with kidney deficiency constitution have changed,indicating that the mother mice with kidney deficiency constitution have an effect on the formation of kidney deficiency constitution and the level of immunoglobulin in offspring mice.
