葛根素在眼用微透析探针上的回收率研究 被引量:3

摘要 目的考察葛根素在自制眼用线性微透析探针上的体内外回收率与灌流速度、药物浓度的关系,以期为葛根素在兔眼内的药动学研究提供依据。方法采用HPLC测定葛根素浓度,通过体外正向法、反向法与体内反向法研究不同灌流速度和药物浓度对微透析探针回收率的影响。结果在所研究的药物浓度和灌流速度的范围内,葛根素在微透析探针上的回收率与药物浓度无关,与灌流速度成反比;体外正、反向回收率之间在灌流速度为1.0、2.0和3.0μL·min^(-1)时均无显著性差异(P>0.05);体内反向回收率显著低于体外正、反向回收率(P<0.05)。结论葛根素在自制线性微透析探针上的回收率稳定,微透析技术可用于葛根素的眼部药代动力学研究。 OBJECTIVE To investigate the relationship between the recovery of puerarin on self-made linear microdialysis probes and the perfusion speed and drug concentrations for further ocular pharmacokinetic study. METHODS The concentration of puerarin in microdialysate was determined by HPLC. The in vitro recovery of puerarin was studied using positive dialysis and retrodialysis methods at different flow rates and concentrations, respectively. And the in vivo recovery of puerarin was studied using retrodialysis methods. RESULTS Within the range of drug concentration and perfusion rate studied, the recovery rate of puerarin on microdialysis probe was independent of drug concentration and inversely proportional to perfusion rate. There was no significant difference between the positive dialysis and retrodialysis recovery in vitro at the perfusion flow rate of 1.0、2.0 or 3.0 μL·min^(-1)(P>0.05), the retrodialysis recovery in vivo was significantly lower than those of in vitro(P<0.05). CONCLUSION The recovery rate of the self-made linear probe is stable, and the microdialysis technique can be used for the study of the ocular pharmacokinetics of puerarin.
