摘要
对一株适用于采油的枯草芽孢杆菌发酵培养基组成和发酵条件进行了优化。通过实验室内的摇床发酵,得到优化后培养基为:蔗糖50 g/L,NaNO_(3)3.4 g/L,NH_(4)Cl 1.1 g,KH_(2)PO_(4)2.5 g/L,12H_(2)O·Na_(2)HPO_(4)30 g/L,7H_(2)O·MgSO_(4)0.8 g/L,脂肽平均产量为4.2 g/L,表面张力可降低至25.15 mN/m;同时,因为在采油工业中,每吨NH_(4)Cl比每吨NaNO_(3)的成本低30%左右,所以通过NH_(4)Cl的加入,减少了NaNO_(3)的用量,大大降低了发酵成本;最佳培养条件为:温度37℃,装液量150 mL/250 mL,发酵时间100 h。
The fermentation medium composition and fermentation conditions of a strain of Bacillus subtilis suitable for oil recovery were optimized.Through the shaker fermentation in the laboratory,the optimal medium of Bacillus subtilis were:sucrose 50 g/L,NaNO_(3)3.4 g/L,NH_(4)Cl 1.1 g,KH_(2)PO_(4)2.5 g/L,12H_(2)O·Na_(2)HPO_(4)30 g/L,7H_(2)O·MgSO_(4)0.8 g/L.The yield of lipopeptide was 4.2 g/L,and the surface tension was reduced to 25.15 mN/m;at the same time,because in the oil production industry,the cost per ton of NH_(4)Cl is about 30%lower than that of NaNO_(3)per ton,the addition of NH_(4)Cl reduces the dosage of NaNO_(3)and greatly reduces the fermentation cost.The optimal culture conditions were as follows:temperature 37℃,filling volume 150 mL/250 mL,fermentation time 100 h.
