Study on quality standard of Yinqin Mixture

摘要 目的建立茵芩合剂的质量控制方法。方法采用微乳薄层色谱法对制剂中的茵陈、黄芩和大黄进行鉴别;采用高效液相色谱法同时测定绿原酸、咖啡酸、黄芩苷和黄芩素的含量,色谱柱为Diamonsil C_(18)(2)(250 mm×4.6 mm,5μm),流动相为乙腈-0.25%磷酸溶液,梯度洗脱,流速为1.0 ml/min,检测波长为310 nm,柱温为30℃。结果微乳薄层色谱显色清晰且阴性对照无干扰;绿原酸、咖啡酸、黄芩苷和黄芩素分别在5.96~59.60μg/ml(r=0.9991)、0.60~6.00μg/ml(r=0.9992)、40.80~408.00μg/ml(r=0.9993)、1.98~19.84μg/ml(r=0.9994)范围内线性关系良好;平均加样回收率分别为96.56%、101.84%、98.77%和98.98%。结论本法操作简便、结果准确,重复性好,可用于茵芩合剂的质量控制。 Objective To develop a quality control method for Yinqin Mixture.Methods Artemisiae Scopariae Herba,Scutellareae Radix,and Rhei Radix et Rhizoma.were identified by microemulsion thin layer chromatography.Chlorogenic acid,caffeic acid,baicalin,and baicalein were simultaneously determined by high performance liquid chromatography,chromatographic column was Diamonsil C_(18)(2)column(250 mm×4.6 mm,5μm),the mobile phase consisted of acetonitrile-0.25%phosphoric acid solution with gradient elution at a flow rate of 1.0 ml/min,the detection wavelength was 310 nm and the column temperature was 30℃.Results The color of microemulsion thin layer chromatography was clear and the negative had no interference;the linear range of chlorogenic acid,caffeic acid,baicalin,and baicalein were 5.96-59.60μg/ml(r=0.9991),0.60-6.00μg/ml(r=0.9992),40.80-408.00μg/ml(r=0.9993),1.98-19.84μg/ml(r=0.9994),respectively;average recovery rates of components were 96.56%,101.84%,98.77%,and 98.98%,respectively.Conclusion The method is simple,accurate,good reproducibility,and can be used for the quality control of Yinqin mixture.
