摘要
介绍研究中药肝毒性的重要性及其现状,中药毒性古今不同定义和现代研究对其毒性成分和作用机制的研究情况,以及系统生物学技术在中药毒性研究中的优势,系统生物学各方面的技术应用在中药毒理学当中的研究进展,包括通过对不同基因比较的基因组学分析、通过进行比较差异蛋白表达的蛋白组学分析、通过差异代谢物的代谢组学比对分析以及通过基因转录和基因表达差异的转录组学分析等现代技术手段进行的更加广泛和深入的中药肝毒性机制研究,此类新型的中药肝毒性研究思路和方法既与中医的整体观念更加接近,而且也使得中药毒性研究手段上升到更加先进的水平,开辟中药毒性研究的新领域,同时还有望使人们对中药肝毒性的认识取得更多突破性进展。
This paper introduces the importance and status quo of researches on hepatotoxicity of traditional Chinese medicine(TCM),the different definitions of toxicity of TCM in ancient and modern times,the research situation of its toxic components and action mechanism,the advantages of systems biotechnology in toxicity research of TCM,and the research progress of the application of systems biotechnology in toxicology of TCM,which includes a more extensive and in-depth study on hepatotoxicity mechanism of TCM by modern technical means such as comparative genomics analysis of different genes,proteomics analysis of comparative differential protein expression,comparative metabonomics analysis of differential metabolites,and transcriptomics analysis of gene transcription and differential gene expression.This new thinking and method of research on hepatotoxicity of TCM is not only closer to the concept of holism of TCM,but also makes the research methods of toxicity of TCM rise to a more advanced level,breaking new ground of research on toxicity of TCM,and at the same time,it is expected to make more breakthrough progress in people's understanding of hepatotoxicity of TCM.
