基于对BMP7的调控探讨MEBO对糖尿病创面愈合的影响 被引量:2

摘要 目的通过湿润烧伤膏(moist exposed burn ointment,MEBO)对骨形态发生蛋白7(bone morphogenetic proteins 7,BMP-7)的调控来探讨其对糖尿病创面愈合的影响。方法按照随机数表将75只Wistar雄性大鼠分为空白组(n=15)、对照组(n=15)、模型组(n=15)、重组牛碱性成纤维细胞生长因子(贝复济)组(n=15)以及MEBO组(n=15),对模型组、贝复济组及MEBO组构建糖尿病模型。生理盐水外敷对照组及模型组创面,MEBO组和贝复济组创面分别给予MEBO、贝复济外敷。各组于治疗后的第1、5、11、18、25天观察创面大小变化及愈合情况,并于上述各个时间点各组随机取3只大鼠在麻醉后快速取创面组织并处死大鼠,标本行HE染色观察创面病理变化及免疫荧光法检测BMP-7的表达。结果(1)在治疗过程中,对照组、贝复济组、MEBO组的病理形态明显改善。(2)BMP-7免疫荧光的变化:除了空白组,其他四组的阳染细胞均升高后降低,具体为第5天出现阳染细胞数的高峰是对照组、贝复济组和MEBO组,而第18天达到峰值的是模型组。(3)在模型组、对照组及MEBO组中,BMP-7表达有差异(P<0.05);而在贝复济组与MEBO组中无差异表达(P>0.05)。结论在糖尿病创面组织中,MEBO可能通过调控BMP-7的表达从而促进溃疡创面的愈合。 Objective To investigate the effect of moist exposed burn ointment(MEBO)on diabetic wound healing by analyzing its regulation on bone morphogenetic proteins 7(BMP-7).Methods According to random number table method,75 male Wistar rats were divided into blank group(n=15),control group(n=15),model group(n=15),recombinant bovine basic fibroblast grow(Beifuji)group(n=15)and MEBO group(n=15).Diabetes models were established in the model group,the Beifuji group and the MEBO group,and the wounds of the model group and the control group were supplied with normal saline,and the wounds of the Beifuji group and the MEBO group were supplied with Beifuji and MEBO,respectively.The changes of wound size and healing were observed on the 1st,5th,11th,18th and 25th days after treatment,and 3 rats in each group were randomly selected at the above time points.After anesthesia,the wound tissue was quickly taken and the rats were killed.The pathological changes of wound were observed by HE staining and the expression of BMP-7 was detected by immunofluorescence.Results(1)During treatment,the pathological morphology of the control group,the Beifuji group and the MEBO group considerably improved.(2)Changes of BMP-7 immunofluorescence:except the blank group,the positive cells of the other four groups increased and then decreased.Specifically,the peak of the number of positive cells on the 5th day of treatment appeared in the control group,the Beifuji group and the MEBO group,while the model group reached the peak on the 18th day of treatment.(3)There were differences in the expressions of BMP-7 among the model group,the control group and the MEBO group(P<0.05);however,there were no differences in expressions between the Beifuji group and the MEBO group(P>0.05).Conclusion In diabetic wound tissues,MEBO may promote the healing of ulcer wounds by regulating the expression of BMP-7.
