摘要
为了查明云南省昆明市郊某规模猪场保育猪群持续发生腹泻的病因,本试验开展了猪主要腹泻病毒核酸的RT-PCR检测及病原菌的分离、培养和鉴定。结果显示,猪主要腹泻病毒核酸检测均为阴性,但置于37℃5%、CO_(2)培养条件下的血清琼脂平板上生长出无色至淡黄色、圆形、凸起、光滑、边缘整齐的菌落,而厌氧培养条件下的血清琼脂平板上无任何菌落生长。分离菌株经革兰染色镜检呈革兰阴性短杆菌,细菌生化鉴定(ID 32E试条)及16S rRNA序列分析均表明该分离菌株为摩氏摩根菌(Morganella morganii)。该分离菌株的16S rRNA序列与NCBI GenBank中的摩氏摩根菌种SDTA-2株(MH299416.1)和FDAARGOS-172株(CP014026.2)的16S rRNA核苷酸序列同源性达到99.78%。昆明小鼠致病性试验显示,该菌株具有一定致病力,致死率为60%(6/10)。细菌药敏鉴定(ATB VET试条)显示,该菌株对青霉素、阿莫西林、夫西地酸、四环素、头孢噻吩等试条上的所有28种药物均耐药,提示该菌株耐药谱较广。本试验为诊治摩氏摩根菌感染导致的仔猪腹泻提供有益的临床及理论参考。
In order to confirm the cause of diarrhea in piglets that persisted in a large scale pig farm in Kunming suburb,this study employed nucleic acid tests to detect major porcine diarrhea-causing viruses and in vitro culture to isolate and identify culprit pathogenic bacteria.The results showed that the nucleic acid tests for major porcine diarrhea-causing viruses were all negative,while serum agar plate at 37℃and with 5%CO_(2)yielded colorless to yellowish,round,raised,smooth colonies with neat edge but serum agar plate under anaerobic culture condition generated no colony.The isolate was found to be Gram-negative short bacillus,and identified as Morganella morganii by biochemical identification(ID 32 E test strip)and 16 S rRNA sequence analysis.Specifically,the 16 S rRNA sequences of this isolate shared 99.78%homology with those of M.morganii SDTA-2(MH299416.1)and FDAARGOS-172(CP014026.2)strains in NCBI GenBank.The pathogenicity test in Kunming mice showed that the strain produced a lethal rate of 60%(6/10),highlighting its pathogenic potential.The bacteria drug sensitivity identification(ATB VET test strip)showed that the strain was resistant to all 28 drugs on the test strip.This study thus serves as clinical and theoretical reference for the prevention and treatment of diarrhea in piglets caused by M.morganii.
