皱皮木瓜品种的遗传多样性和SSR指纹图谱分析 被引量:2

Analysis of Genetic Diversity and SSR Marker Fingerprinting of Chaenomeles speciosa(Sweet) Nakai

摘要 为研究山东省皱皮木瓜品种间的亲缘关系远近,本研究利用PCR结合毛细管电泳法对60份皱皮木瓜种质资源进行遗传多样性分析及指纹图谱构建。所选用的10对多态性高、带型稳定的引物共扩增出55个多态性位点;观测杂合度和期望杂合度的平均值分别为0.392和0.590;Shannon信息指数和Nei’s遗传多样性指数的平均值分别为1.184和0.585;PIC值在0.238~0.819之间,平均值为0.547;聚类分析结果显示,遗传相似系数变幅为0.55~0.96,说明60份皱皮木瓜品种具有较为丰富的遗传多样性。在遗传相似性系数0.662处可将所有供试材料分为3大类。对60个皱皮木瓜品种进行指纹分析,发现具有特征谱带的品种共19个;采用6对引物组合(Nak2,Nak3,Nak5,Nak6,Nak7和Nak10)的方式构建了能够有效区分60个皱皮木瓜品种的指纹图谱。引物Nak5具有较强的鉴别力,在进行图谱构建及分子鉴定中可优先采用。本研究可为皱皮木瓜的遗传资源管理、良种鉴定提供理论基础,同时为品种选育提供参考依据。 In order to explore the genetic relationship among the germplasm varieties of Chaenomeles speciosa in Shandong Province,the genetic diversity of 60 Chaenomeles speciosa germplasm resources was analyzed by the PCR amplification combined with capillary electrophoresis,and their construct fingerprints were constructed.55 polymorphic loci were detected by 10 pairs of primers with high polymorphism and stable type amplified.The average number of observations of heterozygosity and expected heterozygosity were 0.392 and 0.590 respectively.The average number of Shannon information index and Nei’s genetic diversity index were 1.184 and 0.585 respectively.The PIC ranged from 0.238 to 0.819 with an average of 0.547.The results of clustering showed that the genetic similarity coefficient among 60 Chaenomeles speciosa varieties ranged from 0.55 to 0.96.The above results indicate that the genetic diversity of 60 varieties was higher.60 varieties were divided into 3 categories at the genetic similarity coefficient of 0.662.Fingerprint analysis of 60 Chaenomeles speciosa varieties showed 19 varieties with characteristic bands.Fingerprints of 60 varieties were constructed by using 6 pairs of primer combinations(Nak2,Nak3,Nak5,Nak6,Nak7 and Nak10).Primer Nak5 has strong discrimination,which can be preferred in fingerprinting construction and molecular identification.This study can provide a theoretical basis for genetic resource management and varieties identification of Chaenomeles speciosa resources,and provide reference basis for breeding new varieties.
