实时荧光PCR法检测明胶的牛、猪源性成分 被引量:1

摘要 为了快速准确鉴别明胶的动物源性,根据线粒体DNA cytochrome c oxidase subunit I(COX I)基因的种间多态性差异合成特异性引物及探针,运用实时荧光PCR技术扩增与识别特异性基因片段,建立明胶牛、猪源性成分的荧光PCR检测方法。该方法特异性强、灵敏度高、重复性较好,且操作简便,对掺杂牛源性或猪源性成分的检出限可达到0.1%,克服了明胶黏度高、不易溶解,多重工艺对检测结果干扰严重的难点,为明胶的动物源性成分检测提供了有效手段,保障食品添加剂的品质与安全。 In order to quickly and accurately identify the animal origin of gelatin,primers and probes were synthesized based on the characteristic sequence of cytochrome coxidase subunit I(COX I)in the mitochondrial DNA of interspecies.Real-time fluorescent PCR technology was used to amplify and identify specific gene fragments and establish the detection method of bovine and porcine derived materials.The method has the advantages of strong specificity,high sensitivity and repeatability,and with simple operation.The detection limit of doped bovine and porcine derived materials can reach 0.1%.This method overcomes the difficulty of high viscosity and insolubility,and multi-process interference to the test results.It provides an effective means for the detection of animal-derived components of gelatin,and ensures the quality and safety of food additives.
