摘要
G-四链体(G-quadruplexe,G4s)是富含鸟嘌呤DNA/RNA序列折叠形成的稳定分子内或分子间核酸二级结构,受核酸序列、核酸链数量、链的方向、环的位置以及介质环境因素影响而表现出不同的拓扑结构。经过总结文献发现,G4s广泛存在于不同生物染色体中,并在原核和真核生物中具有一定的分布规律,在细胞某些生理过程中发挥着重要的生物学功能。本文从探针种类、测定表征方法等方面对G4s体内、外测定进行了简要说明。此外,G4s的形成因与蛋白质结合而发挥重要作用。G4s与肿瘤、心肌纤维化、肌萎缩侧索硬化症、额颞叶痴呆和白血病等有密切关系。
G-quadruplex(G4s),a stable intra-or inter-molecular nucleic acid secondary structure,is formed by the folding of guanine-rich DNA or RNA sequence.It shows different topological structure under the influence of nucleic acid sequence,nucleic acid chain number,chain direction,ring position and medium environment.After consulting a large number of literatures,it is found that G4s are widely distributed in the chromosomes of different organisms,and have a special distribution rule in prokaryotes and eukaryotes,playing an important biological function in some physiological processes of cells.This paper is an overview for in vivo and in vitro determination methods of G4s,which was briefly described by probe types,determination methods and characterization.Among them,the formation of G4s and protein binding play an important role.G4s are closely related to tumor,myocardial fibrosis,amyotrophic lateral sclerosis,frontotemporal dementia and leukemia.
