摘要
为分析图们江大麻哈鱼Oncorhychus keta生殖洄游群体遗传多样性和遗传结构,评估图们江大麻哈鱼种质资源现状和多年来人工增殖放流对图们江大麻哈鱼群体遗传多样性及遗传结构的影响,利用已发表的10对微卫星引物分析2015、2016、2017及2018年在吉林省图们江流域大麻哈鱼生殖洄游季节采捕的169尾大麻哈鱼亲鱼样本的遗传多样性。分析结果表明,4个年度大麻哈鱼群体遗传多样性水平较高,共检测到等位基因397个,各年度有效等位基因数(Ne)4.7853~6.1309,平均观测杂合度(Ho)0.6500~0.7200,平均期望杂合度(He)0.7663~0.8359,平均多态信息含量(PIC)0.7288~0.8017,平均香农信息指数(I)1.8128~1.9836。变异主要存在于各年度群体内,各年度之间的分化系数为7.55%(Fst=0.0755)。本研究认为,图们江大麻哈鱼生殖洄游群体遗传多样性较丰富,受逐年人工增殖放流的影响不大,适宜继续人工增殖放流来恢复大麻哈鱼资源量。本研究结果可为图们江大麻哈鱼人工增殖放流效果评估提供了参考依据。
The genetic diversity of chum salmon(Oncorhychus keta)was analyzed in 169 individuals collected from the homing chum salmon in Tumen River in 2015, 2016, 2017 and 2018 by 10 published pairs of microsatellite primers to evaluate the genetic diversity and populations structure of the homing chum salmon and current germplasm situation, and to examine effects of enhancement release on the genetic diversity in Tumen River. The analysis showed that there was high genetic diversity in the four annual populations, and a total of 397 alleles were detected in all the 169 individuals, with effective number of alleles ranging from 4.7853 to 6.1309. The average observed heterozygosity was found to be ranged from 0.6500 to 0.7200, the average expected heterozygosity from 0.7663 to0.8359;the average polymorphism information content ranged from 0.7288 to 0.8017, and the average Shannon’s information index ranged from 1.8128 to 1.9836. The variation was mainly found in the annual population, and only 7.55% of the differentiation coefficient between years(Fst=0.0755). It can be seen that the homing chum salmon in Tumen River is rich in genetic diversity, which is not affected by enhancement release, thus it is suitable to continue the enhancement release to recover the salmon resources. The findings provide reference with assessment of the effect of enhancement release of chum salmon in Tumen River.
