摘要
目的探究GSPE对精索扭转损伤大鼠睾丸组织结构、生精细胞的影响。方法把实验大鼠平均分为3组,sham组、TT组和GSPE组。测定三组大鼠氧化应激指标,包括SOD活性和MDA含量;测量大鼠睾丸质量和Johnsen评分;观察睾丸组织形态结构的改变及生精细胞的凋亡指数;免疫组化检测睾丸组织中Bax和Bcl-2的表达。结果和sham组相比,TT组SOD活性降低,但MDA含量升高(P<0.05);干预后的GSPE组大鼠睾丸组织中SOD活性升高,MDA含量得到抑制(P<0.05)。TT组大鼠左侧睾丸组织质量轻于sham组,且Johnsen评分也显著降低(P<0.05);干预后的GSPE组大鼠左侧的睾丸质量显著增加,Johnsen评分也高于TT组(P<0.05)。TT组较sham组大鼠生精小管内的细胞层数减少,细胞松散无结构,形成大量空洞;该组大鼠的生精细胞凋亡指数也显著增加(P<0.05);和TT组相比,GSPE组大鼠睾丸组织结构接近正常,且大鼠生精细胞的凋亡指数显著降低(P<0.05)。免疫组化结果显示,和sham组相比较,TT组大鼠睾丸组织中Bax表达显著升高,Bcl-2显著降低(P<0.05);和TT组相比,GSPE组睾丸组织中Bax表达降低,Bcl-2表达升高(P<0.05)。结论GSPE可改善精索扭转损伤后睾丸的组织结构,并有效抑制生精细胞凋亡,进而对睾丸进行保护,其作用机制与调控Bax/Bcl-2及改善氧化应激水平有关。
Objective To investigate the effects of GSPE on testicular tissue structure and spermatogenic cells in rats with Torsion of spermatic cord injury.Methods The rats were divided into three groups,sham group,TT group and GSPE group.The oxidative stress indexes,including SOD activity and MDA content,testicular weight and Johnsen score,morphological changes of testis were observed by H-E staining.Apoptosis index of spermatogenic cells was also detected by TUNEL.The expression of Bax and Bcl-2 in testis was compared by immunohistochemistry.Results Compared with sham group,the activity of SOD in TT group decreased and the content of MDA increased in TT group(P<0.05);after intervention,SOD activity increased and MDA content was inhibited in GSPE group(P<0.05).The weight of left testicular tissue in TT group was lighter than that in sham group,and Johnsen score was significantly decreased(P<0.05);after intervention,the weight of left testis in GSPE group was significantly increased,and the Johnsen score was also higher than that in TT group(P<0.05).Compared with sham group,the number of primary and secondary spermatocytes in seminiferous tubules in TT group was significantly decreased,and the structure was loose and disordered,with cavity formation;the apoptosis index of spermatogenic cells in this group was also significantly increased(P<0.05).Compared with TT group,the testicular tissue structure of GSPE group was close to normal,and the apoptosis index of spermatogenic cells was significantly decreased(P<0.05).The results of immunohistochemistry showed that compared with sham group,the expression of Bax and Bcl-2 in TT group were significantly higher than those in sham group(P<0.05).Compared with TT group,the expression of Bax was decreased and the expression of Bcl-2 was increased in GSPE group(P<0.05).Conclusion GSPE can improve the histological structure of testis after Torsion of spermatic cord injury,and effectively induce apoptosis of spermatogenic cells,thus protecting the testis.The mechanism of GSPE is related to the regulation of Bax/Bcl-2 and the improvement of oxidative stress level.
