芪参还五胶囊中虫类药材的仿生酶解工艺优化及抗凝血、溶栓活性评价 被引量:3

Optimization of Bionic Enzymatic Hydrolysis Process and Evaluation of Anticoagulant and Thrombolytic Activities of Insect Drugs in Qishen Huanwu Capsules

摘要 目的筛选并优化芪参还五胶囊中虫类药材的仿生酶解工艺条件,并对其抗凝血、溶栓活性进行初步评价。方法以总蛋白含量、活化部分凝血活酶时间(APTT)、凝血酶原时间(PT)、血浆凝血酶时间(TT)和溶栓活性为指标,评价水煎煮、水提醇沉、匀浆、胃蛋白酶酶解、胰蛋白酶酶解和仿生酶解等不同提取方法对芪参还五胶囊中虫类药材的提取效果,并采用正交试验设计优化仿生酶解提取工艺的最佳工艺条件。结果仿生酶解提取物的总蛋白含量、APTT、PT、TT和溶栓活性均为最佳,血栓抑制率为41.97%,与阿司匹林阳性对照的抗凝血和溶栓作用相当。正交试验结果表明,加酶量和酶解时间两个因素对仿生酶解法提取效果有显著性影响(P<0.05),综合得出最佳仿生酶解工艺条件为药材粉碎粒度为80目,加酶量为2%生药量,胃蛋白酶和胰蛋白酶各自酶解时间为2 h。3次平行工艺验证结果表明该工艺条件稳定可行。结论本研究筛选的仿生酶解法工艺条件稳定可行,可作为芪参还五胶囊中三味虫类药材的最佳提取方法。 Objective To screen and optimize the bionic enzymatic hydrolysis process of insect drugs in Qishen Huanwu Capsules and to preliminarily evaluate the anticoagulant and thrombolytic activities.Methods Total protein content,activated partial thromboplastin time(APTT),prothrombin time(PT),plasma thrombin time(TT)and thrombolytic activity were used as indexes to evaluate the extraction effects of different extraction methods,such as water decocting,water extraction and alcohol precipitation,homogenization,pepsin enzymolysis,trypsin enzymolysis and bionic enzymolysis,on insect drugs in Qishen Huanwu Capsules.Orthogonal experiment was used to optimize the optimum conditions of the extraction process of bionic enzymatic hydrolysis.Results The total protein content,APTT,PT,TT and thrombolytic activity of the biomimetic enzymatic extract were best,and the thrombus inhibition rate was 41.97%,and the anticoagulant and thrombolytic effects were similar to those of aspirin.The results of orthogonal test showed that the amount of the enzyme and hydrolysis time had significant effects on the extraction efficiency of biomimetic enzymatic hydrolysis(P<0.05).In conclusion,the optimal conditions of bionic enzymatic hydrolysis were as follows:the medicinal material powders were screened through an 80 mesh sieve,the amount of the enzyme was 2%of the crude drug,and the enzymatic hydrolysis time of pepsin and trypsin was 2 h,respectively.The results of three parallel validation experiments showed that the process condition was stable and feasible.Conclusion The bionic enzymatic hydrolysis process screened out in this study is stable and feasible,and can be used as the best extraction method of insect drugs in Qishen Huanwu Capsules.
