摘要
为了观察猪红细胞与猪肺泡巨噬细胞(PAMs)相互作用以清除免疫复合物的生物学过程,采用显微镜技术和流式细胞技术,借助平行板流动小室灌流系统构建猪红细胞免疫黏附致敏GFP-Escherichia coli与PAMs的细胞动态互作模型,并对GFP-Escherichia coli从猪红细胞表面向PAMs转移的过程进行了研究。结果表明,黏附有GFP-Escherichia coli的猪红细胞流动至PAMs处停留,绿色荧光点位于红细胞与PAMs交接处。随后,猪红细胞脱离连接,GFP-Escherichia coli完全转移至PAMs并被PAMs吞噬清除。试验成功拍摄到PAMs移除猪红细胞表面GFP-Escherichia coli的过程,可为猪红细胞免疫黏附介导免疫复合物清除的研究提供可视化的证据。
To observe the biological process of porcine erythrocytes interacting with porcine alveolar macrophages(PAMs)to clear immune complexes,microscopy and flow cytometry were utilized to construct a dynamic interaction model of porcine erythrocytes immune-adhesion opsonized GFP-Escherichia coli and PAMs with the assistance of a parallel plate flow cell perfusion system.The transfer process of GFP-Escherichia coli from the surface of porcine erythrocytes to PAMs was observed by fluorescence microscope.The results showed that a porcine erythrocyte adhered with GFP-Escherichia coli moved to the PAMs and stayed there.The green fluorescent spot was located at the junction of the porcine erythrocyte and PAMs.Subsequently,the porcine erythrocyte was disconnected,and GFP-Escherichia coli was completely transferred to PAMs and was swallowed by PAMs.This experiment successfully captured the removal of GFP-Escherichia coli from the surface of porcine erythrocytes by PAMs,which could provided visualized evidence of porcine erythrocyte immune adhesion-mediated immune complex clearance.
