利用LC-MS/MS辅助优化GICA对硝基呋喃类代谢物的快速检测 被引量:2

摘要 针对硝基呋喃类代谢物的胶体金免疫层析法(GICA)检测方法存在的缺陷,实验以硝基呋喃类代谢物实际阳性样本和模拟阳性样本为研究对象,选择液相色谱-串联质谱(LC-MS/MS)辅助优化GICA代谢物的水解和衍生条件前处理步骤,建立了一种快速检测水产品中硝基呋喃类代谢物的方法。结果显示最佳实验条件为:盐酸浓度0.35 mol/L,0.05 mol/L 2-硝基苯甲醛0.35 mL,水浴温度90℃,水浴时间120 min。此条件下,胶体金免疫层析法的前处理时间缩短至120 min,检测时间为5 min,4种硝基呋喃类代谢物的检出限均为0.05μg/kg且重现性好。本方法对140批次不同基质样品进行分析,检测得出12批次阳性样品,假阳性和假阴性率均为0。结果表明本方法在降低实验前处理时间同时,适用于不同的样品基质,可应用于大批量实际样品的测定。 In view of the drawback of GICA detection method for nitrofuran metabolites,this study developed a rapid detection method for nitrofuran metabolites in aquatic products.The actual positive samples and simulated positive samples of nitrofuran metabolites were selected as research objects,and the liquid chromatography-tandem mass spectrometry(LC-MS/MS)was applied as the auxiliary for the pretreatment steps of the hydrolysis and derivatization conditions of the colloidal gold immunochromatography(GICA)metabolites.The results showed that the optimal experimental conditions were:hydrochloric acid concentration of 0.35 mol/L,2-nitrobenzaldehyde(0.05 mol/L)of 0.35 mL,water bath temperature of 90℃and water bath time of 120 min.Under these conditions,the pretreatment time of colloidal gold immunochromatography was shortened to 120 min,and the detection time was 5 min.The detection limits of four nitrofuran metabolites were all 0.05μg/kg,along with good reproducibility.Different matrix samples of 140 batches were analyzed by this developed method,among which positive samples of 12 batches were detected,and both false positive and false negative rates were 0.The results indicated that this method was suitable for different matrix samples while reducing the pre-processing time of the experiment,and it could be applied to the determination of actual samples with large quantities.
