苦笋壳多糖提取工艺及抗氧化活性研究 被引量:14

Extraction Optimization for Polysaccharides from Bamboo Shoot Shell of Pleioblastus Amarus and the in Vitro Antioxidant Activity

摘要 目的:确定苦笋壳多糖的最佳提取工艺,并对其抗氧化活性进行研究。方法:通过考察料液比、酶用量、酶解时间、酶解温度、酶解pH值对苦笋壳多糖得率的影响,在单因素实验基础上采用Box-Behnken响应面法建立数学模型,优化最佳提取工艺条件并扫描电镜观察苦笋壳多糖形态。以DPPH、ABTS、·OH和O_(2)^(-)·清除能力、FRAP还原能力为指标,考察其体外抗氧化活性。结果:回归模型差异显著(P<0.05),可对苦笋壳多糖含量进行很好地分析和预测;得到最佳工艺参数:料液比1:35、酶用量3.11%、酶解时间101 min、酶解温度54℃、pH7.0,此条件下模型预测的最大提取量为25.7 mg/g,实测值为(25.4±0.1420)mg/g,与预测值无显著性差异。该多糖为不规则形态,具有较多的分枝,表面平整光滑,边缘呈锯齿状。其DPPH、ABTS、·OH和O_(2)^(-)·的IC50分别为2.47、2.12、0.2、13.58 mg/mL,FRAP还原能力随多糖浓度的增加而上升。结论:由响应面法优化得到的苦笋壳多糖的提取工艺方便、可行,得到的苦笋壳多糖具有较强的抗氧化活性。 Objective:This study aimed to determine the optimum extraction technology of polysaccharides from bamboo shoot shell of Pleioblastus amarus a nd s tudy i ts a ntioxidant a ctivity.M ethods:B y investigating the effects of feed-to-liquid ratio,enzyme dosage,enzymatic hydrolysis time,enzymatic hydrolysis temperature and enzymatic hydrolysis pH on the yield of bamboo shoot shell polysaccharide of Pleioblastus amarus,a mathematical model was established based on the single factor experiment using Box-Behnken response surface method to optimize the optimal extraction conditions.The scanning electron microscope(SEM)were used to study the structure of crude polysaccharide.The DPPH,ABTS,·OH and O_(2)^(-)·scavenging ability and FRAP reducing ability were used as indicators to investigate the in vitro antioxidant activity.Results:The difference of the regression model was significant(P<0.05).The optimum extraction conditions were as follows:the ratio of material and liquid was 1:35,the amount of enzyme was 3.11%,the time of enzymolysis was 101 min,the temperature of enzymolysis was 54℃,and pH 7.0.Under this condition,the maximum extraction amount of the model was 25.7 mg/g,and the measured value was(25.4±0.1420)mg/g,which had no significant difference from the predicted value.The surface structure of crude polysaccharide showed a irregular structure with many branches,and the surface was smooth and the edge was serrated.The IC50 of DPPH,ABTS,·OH and O_(2)^(-)·were 2.47 mg/mL,2.12 mg/mL,0.2 mg/mL,13.58 mg/mL.The reduction ability of FRAP increased with the increase of polysaccharide concentration.Conclusion:The extraction process of the polysaccharides from the bamboo shoot shell of Pleioblastus amarus optimized by the response surface method is convenient and feasible.And the polysaccharides has strong antioxidant activity.
