三七皂苷对T2DM大鼠胰岛素抵抗的影响 被引量:5

摘要 目的分析三七皂苷对2型糖尿病(T2DM)大鼠胰岛素抵抗的影响。方法选取清洁级Wis-tar大鼠60只,采用简单随机数字表法随机分为正常组(蒸馏水10 ml/kg)、模型组(蒸馏水10 ml/kg)、低剂量组(50 mg/kg三七皂苷)、中剂量组(100 mg/kg三七皂苷)、高剂量组(200 mg/kg三七皂苷)、二甲双胍组(100 mg/kg二甲双胍)各10只,采用链脲佐菌素(STZ)结合高脂饲料喂养法建立T2DM大鼠模型,连续给药4周后取胰腺组织进行苏木精-伊红(HE)染色,并计算胰腺β细胞凋亡指数(AI),测定空腹血糖(FPG)、糖化血红蛋白(HbA1c)、胰岛素(FINS)、胰岛素抵抗指数(IRI)、胰岛素敏感指数(ISI)及肝脏组织肿瘤坏死因子-α(TNF-α)。结果三七皂苷各组的胰岛β细胞形态、纤维组织破坏、腺泡与胰岛散在交错较模型组减少;模型组AI高于正常组及三七皂苷各组、二甲双胍组,其中三七皂苷高剂量组、二甲双胍组AI均低于低剂量组、中剂量组(P<0.05);正常组FPG、HbA1c、FINS、IRI低于其他各组,而ISI高于其他各组,模型组FPG、HbA1c、FINS、IRI高于三七皂苷各组、二甲双胍组,ISI低于三七皂苷各组、二甲双胍组,其中高剂量组差异最明显(P<0.05),高剂量组各指标与二甲双胍组比较差异无统计学意义(P>0.05);各组肝脏组织TNF-α比较差异有统计学意义(P<0.05),其中模型组TNF-α最高,其次为低剂量组,高剂量组TNF-α与二甲双胍组比较差异无统计学意义(P>0.05)。结论三七皂苷可改善T2DM大鼠的糖代谢紊乱及胰岛素抵抗,可能与其增加胰岛素敏感性、抑制肝脏组织炎性因子TNF-α释放等有关。 Objective To determine the effect of notoginsenoside on insulin resistance in type 2 diabetes mellitus(T2DM)rats.Methods Sixty clean Wistar rats were included and randomly divided into normal group(10 mL/kg distilled water),model group(10 mL/kg distilled water),low-dose group(50 mg/kg notoginsenoside),medium-dose group(100 mg/kg notoginsenoside),high-dose group(200 mg/kg notoginsenoside),and Metformin group(100 mg/kg Metformin),with 10 rats in each group.Streptozotocin(STZ)combined with high-fat diet feeding method was used to establish T2DM rat model.At four weeks after continuous administration,pancreatic tissues were harvested for HE staining,and pancreaticβ-cell apoptosis index(AI)was calculated.Fasting plasma glucose(FPG),glycosylated hemoglobin(HbA1c),fasting insulin(FINS),insulin resistance index(IRI),insulin sensitivity index(ISI)and liver tumor necrosis factor-α(TNF-α)were determined.Results Compared with the model group,the morphology of pancreaticβcells,fibrous tissue destruction,and scattered acinar and pancreatic islet in the three notoginsenoside groups decreased.The AI in the model group was higher than that in each notoginsenoside group and Metformin group,and the AI in high-dose group and Metformin group was lower than thatin low-dose group and medium-dose group(all P<0.05).The levels of FPG,HbA1c,FINS,and IRI in normal group were lower than those in the other groups,whereas the level of ISI was higher than that in the other groups.The levels of FPG,HbA1c,FINS and IRI in the model group were higherthan those in thenotoginsenoside groups and Metformingroup,whereas the level of ISI was lower than that inthenotoginsenoside groups and Metformin group.Compared with the high-dose group,the differences were the most significant(all P<0.05).There were no statistically significant differences in the indexes between the high-dose group and the Metformin group(all P>0.05).There was statistically significant difference in TNF-αlevel of liver tissue among the groups(P<0.05),and the TNF-αlevel in the model group was the highest,followed by the low-dose group.There was no statistically significant difference in the TNF-αlevelbetween the high-dose group and Metformin group(P>0.05).Conclusion Notoginsenosidemay improve glucose metabolism disorder and insulin resistance in T2DM rats,which may be related to the increase of insulin sensitivity and inhibition of the release of inflammatory factor TNF-αin liver tissues.
