脂多糖调控肿瘤微环境巨噬细胞变化且抑制胰腺癌发生发展 被引量:2

Lipopolysaccharide regulates macrophages in the tumor microenvironment and inhibits the initiation and development of pancreatic cancer

摘要 目的观察脂多糖(LPS)对胰腺癌小鼠肿瘤生长速度、体积及重量的影响,并探讨机制。方法选取12只健康雌性C57/BL6小鼠,制备小鼠胰腺癌皮下瘤模型,以随机数法分为磷酸盐缓冲液(PBS)组和LPS组,每组6只,每2 d腹腔注射LPS/PBS。比较两组小鼠肿瘤的生长速度、体积及重量,流式细胞术检测肿瘤组织巨噬细胞比例的变化,免疫组织化学检测肿瘤组织巨噬细胞的变化。计量资料间比较采用两独立样本t检验。结果与PBS组比较,LPS组能够显著抑制小鼠肿瘤生长、减小肿瘤体积,最后一次测量小鼠肿瘤体积LPS组较PBS组明显减少[(422.87±129.82)mm^(3)比(2763.91±617.41)mm^(3),F=4.310,t=-7.421,P<0.01],LPS组小鼠肿瘤重量亦显著低于PBS组[(0.22±0.08)g比(0.77±0.16)g,F=1.392,t=-7.480,P<0.01]。流式细胞术结果表明,与PBS组比较,LPS组肿瘤内M1型巨噬细胞比例并无显著升高[(12.00±1.99)%比(10.33±0.74)%,F=5.477,t=1.359,P>0.05],而LPS组M2型巨噬细胞比例较PBS组明显减少[(5.59±0.14)%比(6.60±0.42)%,F=4.424,t=-3.953,P<0.05]。免疫组化结果表明LPS组肿瘤组织内M1型巨噬细胞比例较PBS组差异无统计学意义[(3.0±0.58)/10 HPF比(1.3±0.33)/10 HPF,F=0.400,t=2.500,P>0.05],而LPS组M2型巨噬细胞比例较PBS组明显减少[(7.30±0.33)/10 HPF比(28.30±1.86)/10 HPF,F=8.526,t=-11.137,P<0.01]。结论LPS可抑制胰腺癌小鼠肿瘤生长,发挥抗肿瘤功能,其机制可能与M2型巨噬细胞的减少有关。 Objective To observe the effect of lipopolysaccharide(LPS)on tumor growth and volume in tumor bearing mice of pancreatic cancer and explore the potential mechanism.Methods Twelve female C57/BL6 mice were selected to establish murine subcutaneously pancreatic tumor model,then randomly divided into phosphate buffer saline(PBS)group and LPS group,with 6 mice in each group.The same volume of LPS/PBS was injected intraperitoneally every 2 days.The tumor growth rate,volume and weight of two groups were measured.Flow cytometry was used to analyze differences in the proportions of macrophages within tumor,and immunohistochemistry was used to detect the changes of macrophages in tumor tissue.Results Compared with PBS,LPS could significantly attenuate the growth rate of tumors,reduce tumor volume and weight.Tumor volume of mice measured last time in LPS grou was(422.87±129.82)mm^(3) and that in PBS group(2763.91±617.41)mm^(3) (P<0.01).The tumor weight of mice in the LPS group[(0.22±0.08)g]was also significantly lower than that in the PBS group[(0.77±0.16)g](P<0.01).The results of flow cytometry showed that compared with the PBS group[(10.33±0.74)%],the proportion of M1 macrophages in the tumor of the LPS group[(12.00±1.99)%]did not increase significantly,while the proportion of M2 macrophages was significantly reduced(P<0.05),which was(5.59±0.14)% in LPS group and(6.60±0.42)% in PBS group,respectively.The results of immunohistochemistry showed that there was no significant difference in the proportion of M1 macrophages in tumor(P>0.05),while the proportion of M2 macrophages was significantly reduced(P<0.01).Conclusion LPS can inhibit tumor growth of pancreatic cancer mice model and exert anti-tumor function.The mechanism might be associated with the decrease of M2 macrophages.
